饲用粪肠球菌EXW27培养条件的响应面优化 被引量:3

摘要 本试验旨在通过将单因素、Plackett-Burman筛选与响应面分析3种方法相结合来优化饲用粪肠球菌EXW 27培养条件。以单因素试验为基础,采用Plackett-Burman试验设计筛选显著影响菌株EXW 27发酵活菌数的培养条件,然后用最陡爬坡路径逼近最大响应区域,最后通过响应面分析确定主要影响因素的最佳值。结果表明:菌株EXW 27发酵培养条件为:初始pH 7.7、培养温度38.3℃、装液量120 mL/250 mL、接种量6%、摇床转速210 r/min、培养时间18 h,在此最佳条件下,菌株EXW27的活菌数为3.18×1010 cfu/mL,比优化前提高了近3倍。 The purpose of this experiment was to optimize the culture conditions of Enterococcus faecalis EXW27 by using the methods of single factor and Plackett-Burman(PB)combining with response surface methodology.On the basis of single factor test,Plackett-Burman design was used to screen the fermentation conditions of the significantly affected strain EXW27,and then the path of steepest ascent was adopted to approach the optimal region.Finally,the optimum levels of the main influencing factors was determined by response surface analysis.The results showed that the strain EXW27 fermentation culture conditions as follows:the initial pH 7.7,temperature 38.3℃,volume 120 mL/250 mL,inoculum 6%,shaking speed 210 r/min and culture time 18 h,under the optimum conditions,the count of Enterococcus faecalis EXW27 reach to 3.18×1010 cfu/mL,which was about 3 times than that of before the optimization.
