小尾寒羊MyoG基因外显子1的克隆及多态性分析 被引量:1

摘要 为促进畜禽育种工作的发展,采用测序法和群体遗传学方法进行数据分析,提取12月龄绵羊血液DNA,依据GenBank中羊MyoG基因序列扩增外显子1,研究小尾寒羊肌细胞生成素(Myogenin,MyoG)基因外显子在群体中的多态性。结果表明:在外显子1第211 bp位存在C>T的突变,共得到AA、AB、BB三种基因型。绵羊AA、AB、BB基因型频率分别为0.529,0.353,0.118;等位基因A和B的基因频率分别为0.706和0.294;其中AA基因型频率在0.5以上,为优势基因型。纯合度(Ho)为0.584;杂合度(He)为0.416;有效等位基因(Ne)为1.712;多态信息含量(PIC)为0.330,在0.25~0.50,为中度多态。DNAStar软件对片段同源性对比分析发现其与山羊的同源性为99%,与牦牛的同源性为97%,与猪的同源性为92%。 In order to promote the development of livestock and poultry breeding,data analysis was carried out by sequencing method and population genetics method.Blood DNA of 12-month-old sheep was extracted.MyoG gene exon 1 was amplified,according to GenBank.It was studied the polymorphism of myogenin(MyoG) gene exon in population of Small Tailed Han Sheep.The results showed that C>T mutation at 211 bp in exon 1,and AA,AB and BB genotypes were obtained.The results showed that the genotype frequencies of AA,AB and BB were 0.529,0.353 and 0.118,respectively.The frequency of alleles A and B was 0.706 and 0.294,respectively.The AA genotype frequency was above 0.5,which was the dominant genotype.The homozygosity(Ho) was 0.584;The heterozygosity(He) was 0.416.The effective allele(Ne) was 1.712.Polymorphic information content(PIC) was 0.330 and moderate polymorphic between 0.25 and 0.50.Comparative analysis of fragment homology by DNAStar software showed that the homology of the fragment was 99% with goats,97% with yaks,and 92% with pigs.
