摘要
目的探讨60Co-γ射线辐照灭菌对开胃健脾丸有效成分的影响及辐照前后指纹图谱的变化。方法色谱柱为Agilent Zorbax-C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.5%甲酸溶液,梯度洗脱,检测波长分别为220 nm(检测白术内酯Ⅱ、白术内酯Ⅲ及党参炔苷)、275 nm(检测白术内酯Ⅰ)、300 nm(检测橙皮苷),柱温为30℃,进样量为10μL;建立开胃健脾丸高效液相色谱指纹图谱,同时以0,3,5,8 kGy剂量的60Co-γ射线辐照样品,检测白术内酯Ⅰ、白术内酯Ⅱ、白术内酯Ⅲ、党参炔苷及橙皮苷的含量,以及计算指纹图谱相似度。结果当60Co-γ射线辐照剂量不超过8 kGy时,白术内酯Ⅰ、白术内酯Ⅱ、白术内酯Ⅲ、党参炔苷及橙皮苷的指纹图谱和含量无明显变化。结论60Co-γ射线辐照灭菌法对开胃健脾丸有效成分和指纹图谱无影响,可作为其有效灭菌手段。
Objective To investigate the effect of 60Co-γrays radiation sterilization on the contents of effective constituents and the changes of fingerprint of Kaiweijianpi Pills before and after the radiation sterilization.Methods The chromatographic column was Agilent Zorbax-C18 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.5%formic acid solution with gradient elution,the detection wavelength was set at 220 nm for atractylode Ⅱ,atractylode Ⅲ and lobetyolin,275 nm for atractylodeⅠ,300 nm for hesperidin,the column temperature was 30℃and the sample size was 10μL.The HPLC fingerprint of Kaiweijianpi Pills was established,and the samples were irradiated with 60Co-γrays at the dose of 0,3,5,8 kGy.The contents of atractylolideⅠ,atractylolide Ⅱ,atractylolideⅢ,lobetyolin and hesperidin were detected,and the similarity of the fingerprint was calculated.Results When the dose of60Co-γrays radiation was less than 8 kGy,the fingerprints and contents of atractylolideⅠ,atractylolide Ⅱ,atractylolide Ⅲ,lobetyolin and hesperidin did not change significantly.Conclusion 60Co-γrays radiation sterilization does not affect the effective constituents and fingerprint of Kaiweijianpi Pills,which can be used as an effective sterilization method for Kaiweijianpi Pills.
