川木香中主要药效成分对小鼠急性胃溃疡保护作用的配伍研究 被引量:20

Effects of active components from vladimiriae radix and their combinations on ethanol-induced acute gastric ulcer in mice

摘要 目的:研究川木香中主要药效成分木香烃内酯(CO)、去氢木香内酯(DE)及其不同配伍比例对乙醇诱导的小鼠急性胃溃疡的保护作用。方法:采用无水乙醇复制急性胃溃疡模型,用组织病理学和酶联免疫吸附(ELISA)法评价不同配伍比例对急性胃溃疡的保护作用。结果:CO,DE单用,或不同配伍比例均可抑制炎性因子肿瘤坏死因子-α(TNF-α)、核因子-κB(NF-κB)、一氧化氮(NO)和诱导型一氧化氮合酶(iNOS)以及氧化应激因子丙二醛(MDA)生成,升高超氧化物歧化酶(SOD)和增殖细胞核抗原(PCNA)水平,减轻胃组织病理学损伤等,其中配伍比例为3∶1时对促炎症因子、氧化应激因子和细胞增殖因子的影响最为显著。结论:CO和DE配伍比例为3∶1时,对胃溃疡损伤的保护作用尤为明显,该研究为川木香的进一步研发提供了实验依据。 Objective:To study the protective effects of the main effective components of vladimiriae radix,costunolide(CO),dehydrocostuslactone(DE),and their combinations in different proportions on ethanol-induced acute gastric ulcer in mice.Methods:The acute gastric ulcer model was established with absolute ethanol,then histopathological analysis and enzyme-linked immunosorbent assay(ELISA)were used to evaluate the protective effects of the components and their combinations on acute gastric ulcer.Results:CO and DE alone or their combinations in different proportions could inhibit the production of inflammatory related factors such as tumor necrosis factor-alpha(TNF-α),nuclear factor-κB(NF-κB),nitric oxide(NO),inducible nitric oxide synthase(iNOS)and oxidative stress factor malondialdehyde(MDA),increase the levels of superoxide dismutase(SOD)and proliferating cell nuclear antigen(PCNA),and alleviate the pathological damage of gastric tissue,etc.When the compatibility ratio was 3∶1,it had the most significant effect on proinflammatory factors,oxidative stress factors and cell proliferation factors.Conclusion:When the ratio of CO and DE is 3∶1,the protective effect on gastric ulcer injury is particularly obvious.This study provides experimental basis for further research and development of vladimiriae radix.
