摘要
目的分析孕晚期血浆miRNA表达水平与子痫前期的相关性。方法选择2016年3月至2018年3月北京中医医院顺义医院收治的子痫前期患者83例为观察组,另选取体检的健康妊娠妇女70例作为对照组,测定两组孕妇的收缩压、舒张压及24 h尿蛋白水平,测定血浆miRNA-210基因表达水平,并进行相关性分析。24 h尿蛋白水平采用免疫透射比浊法测定,血浆miRNA-210基因表达水平采用逆转录-聚合酶链法检测,以U 6为内参,检测miRNA-210的相对表达水平。结果观察组子痫前期患者的收缩压、舒张压、24 h尿蛋白水平、血浆miRNA-210表达水平均显著高于对照组,差异有统计学意义(P<0.05)。Spearman相关性分析,子痫前期患者miRNA-126基因表达与收缩压、舒张压、蛋白尿水平均呈正相关(r=0.783、0.826、0.811,P均<0.05)。结论孕晚期血浆miRNA-210表达水平与收缩压、舒张压、24 h尿蛋白水平均有显著相关性,血浆miRNA-210表达水平与子痫前期疾病的发生发展具有较好相关性,血浆miRNA-210有望成为预测子痫前期的潜在标志物。
Objective The correlation between the expression level of plasma miRNA in the third trimester and preeclampsia was analyzed. Methods From March 2016 to March 2018, 83 patients with preeclampsia admitted to Shunyi Hospital of Beijing Traditional Chinese Medicine Hospital were selected as observation subjects. Another 70 healthy pregnant women were selected as the control group. The systolic blood pressure of the two groups was determined. The levels of plasma miRNA-210 gene expression were measured by diastolic blood pressure and 24 h urine protein levels, and correlation analysis was performed. The 24 h urinary protein level was determined by Immunoturbidimetry assay. The expression level of plasma miRNA-210 gene was detected by reverse transcription-polymerase chain method. The relative expression level of miRNA-210 was detected by U 6 as internal reference. Results The systolic blood pressure, diastolic blood pressure, 24 h urine protein level and plasma miRNA-210 expression level in the preeclampsia group were significantly higher than those in the control group(P<0.05). Spearman correlation analysis showed that miRNA-126 gene expression in preeclampsia patients was positively correlated with systolic blood pressure, diastolic blood pressure and urine protein(r=0.783, 0.826, 0.811, all P<0.05). Conclusion The expression level of plasma miRNA-210 in the third trimester is significantly correlated with systolic blood pressure, diastolic blood pressure and 24 h urine protein level. The expression level of plasma miRNA-210 has a good correlation with the occurrence and development of preeclampsia. Plasma miRNA-210 is expected to be a potential marker for predicting preeclampsia.
