摘要
目的 了解佳木斯市禽畜源弯曲菌污染情况及耐药性和分子分型,探讨佳木斯市禽源弯曲菌耐药谱和同源性关系,为食源性疾病溯源和预警提供基础数据。方法 对2018年3-9月来自佳木斯市各类禽畜类销售市场的随机样品85份,应用驱动增强双孔滤膜法分离弯曲菌,经镜检、生化和荧光聚合酶链式反应(PCR)方法进行鉴定,应用琼脂稀释法对细菌进行7大类11种抗菌药物敏感性检测,应用脉冲场凝胶电泳(PFGE)进行基因分型,应用BioNumerics(BN)软件进行聚类分析,比较其同源性。结果 85份样本分离到弯曲菌26株,总分离率为30.59%,全部菌株对萘啶酸抗菌药和环丙沙星耐药率达到100.0%,对四环素抗菌药的耐药率达76.9%,对11种抗菌药物产生了12种耐药谱,42.3%(11/26)出现了多重耐药现象。26株弯曲菌呈现24种PFGE带型,基因型呈现多态性。结论 驱动增强双孔滤膜法可有效分离肉类样本中弯曲杆菌,并且弯曲菌分离株对多种抗菌药物产生了显著的耐药性和多态性,应引起社会各部门的高度重视。
Objective To investigate the contamination,drug resistance and molecular typing characteristics of Campylobacter from poultry and livestock in Jiamusi City,explore the drug resistance spectrum and homology of Campylobacter from poultry,and provide basic data for traceability and early warning of foodbome diseases.Methods 85 random samples were collected from various poultry and livestock markets in Jiamusi City from March to September 2018.Campylobacter was isolated by the driving enhanced dual-pore filtration membrane method,and identified by the microscopic examination,biochemical experiments and fluorescent PCR.Subsequently,sensitivity tests of 11 antimicrobial agents in 7 categories were performed by agar dilution method,and pulsed field gel electrophoresis(PFGE)was used for genotyping.The cluster analysis was performed by BioNunierics(BN)software to compare the homology.Results Of 85 samples,26 were Campylobacter positive,and the total positive rate was 30.59%.The resistance rate of all Campylobacter strains to quinolones and Ciprofloacin reached 100%,and the resistance rate to Tetracyclin was 76.9%.12 kinds of drug resistance spectrum were produced to 11 kinds of antibiotics.42.3%(11/26)of them were multiple drug resistant bacteria.26 Campylobacter strains showed 24 PFGE bands,showing polymorphism of genotype.Conclusion Campylobacter in meat samples can be effectively separated by driving enhanced dual-pore filtration membrane method,moreover,Campylobacter strains show significant resistance and polymorphism to many antibiotics.All sectors of society should attach great importance to it.
