摘要
目的制备临床验方止吐脐贴复方提取物的纳米乳剂型,并考察其理化性质和体外透皮性能。方法利用伪三元相图,对止吐复方纳米乳的处方进行筛选。采用激光粒度仪对纳米乳的粒径与稳定性进行评价。以丁香酚作为中药止吐复方提取物的指标性成分,采用高效液相色谱测定其含量。利用Franz扩散池法小鼠腹部皮肤为渗透皮肤,以含25%乙醇的磷酸盐缓冲液(pH 7.4)溶液作为接收液,进行中药止吐复方纳米乳的体外透皮实验。结果制备的中药止吐复方纳米乳的平均粒径为(26.5±5.6)nm,稳定性好。透皮效果最佳的中药止吐复方纳米乳处方是薄荷油为油相,聚乙二醇-40氢化蓖麻油为乳化剂,卡比醇为助乳化剂,K m=2∶1,混合乳化剂与油相的比例为9∶1。该最优处方在8 h内的累积渗透百分率约为22%,而原剂型的累积渗透百分率仅为2%。结论中药止吐复方纳米乳的透皮给药量相比原剂型有显著提升。
Objective To prepare the nanoemulsion of a clinical prescription,antiemetic compound umbilical paste,and investigate its physicochemical properties in vitro transdermal permeability.Methods The formulation of antiemetic compound nanoemulsion was selected by pseudo-ternary phase diagram.The particle size and stability of nanoemulsion was evaluated by a laser particle size analyzer.Eugenol was utilized as the index component of the antiemetic compound extract,and the content of eugenol was determined by high-performance liquid chromatography.In vitro transdermal experiment of the antiemetic compound nanoemulsion was operated with the methods of Franz diffusion cell.Mouse abdominal skin was used as the skin model and phosphate buffered saline(pH 7.4)containing 25%ethanol was used as the receiving liquid.Results The antiemetic compound nanoemulsion had a mean size of(26.5±5.6)nm and good thermodynamic stability.The optimum formulation of the antiemetic compound nanoemulsion with best transdermal effect was as follows:mint oil as oil phase,polyethylene glycol-40 hydrogenated castor oil as emulsifier,carbitol as co-emulsifier,K m=2∶1,and the mixed emulsifier to oil phase was 9∶1.The cumulative penetration ratio of the optimal prescription within 8 h was ca.22%,while that of the original dosage form was only 2%.Conclusion Comparing with the original dosage form,the transdermal permeability of the antiemetic compound nanoemulsion is improved markedly.
