摘要
目的明确4-硝基喹啉-1-氧化物(4-nitroquinoline 1-oxide,4NQO)诱导C57BL/6鼠食管鳞癌模型中牙龈卟啉单胞菌(Porphyromonas gingivalis,P.gingivalis)对食管鳞癌发生的促进作用。方法120只C57BL/6鼠随机分为对照组、1,2-丙二醇溶剂对照组、4NQO组和4NQO联合P.gingivalis牙周炎组,每组各30只。4NQO诱导食管鳞癌模型中,50μg·mL^-1的4NQO处理8周后,鼠口腔上颌第二磨牙丝线结扎、P.gingivalis口腔持续感染。结果22周时,4NQO组和4NQO联合P.gingivalis组鼠食管黏膜上皮开始发生病变,肉眼观为黏膜表面粗糙,灶性颗粒状或乳头状增生;镜下为食管黏膜基底细胞增生,26周时4NQO联合P.gingivalis组出现不典型增生,36周实验终止时联合组出现癌变。与4NQO单纯处理组比较,P.gingivalis感染组小鼠食管黏膜病变面积明显增大,基底细胞增生和不典型增生发生时间提前。结论P.gingivalis能够促进4NQO诱导C57BL/6鼠食管鳞癌的发生。
Objective To determine the tumorigenic role of Porphyromonas gingivalis(P.gingivalis)in mouse model of 4NQO induced esophageal squamous cell cancer.Methods One-hundred and twenty C57BL/6 mice were randomly and equally assigned to control group,vehicle control group,4NQO group and 4NQO combined with P.gingivalis group.In mouse model of 4NQO induced esophageal squamous cell cancer,mice were fed with 50μg·mL^-14NQO via drinking water for 8 weeks followed by ligature of the maxillary second molar and then oral P.gingivalis infection in the experiment.Results At the 22th weeks,the gross changes in mucosa of mouse esophagus in 4NQO and 4NQO combined with P.gingivalis groups comprised rough surface and focal granular or papillomatous appearance;the microscopic changes consisted of hyper-proliferation of esophageal basal cells;at 26 weeks,dysplasia occurred and at termination of this experiment,esophageal squamous cell cancer occurred in 4NQO combined with P.gingivalis group.The abnormal surface areas,basal cell hyper-proliferation and dysplasia of esophageal mucosa in 4NQO combined with P.gingivalis group occurred earlier than those in 4NQO single treatment group.Conclusion P.gingivalis promotes esophageal squamous carcinogenesis modeled by 4NQO in C57BL/6 mouse.
