摘要
目的:建立慢性非细菌性前列腺炎(chronic nonbacterial prostatitis,CNP)实验动物模型。方法:取40只雄性SD大鼠随机分为4组,除空白对照组外,其他3组分别经腹腔麻醉后行去势手术,术后第二天起多点皮下注射苯甲酸雌二醇0.5、0.3、0.25 mg/kg(即高、中、低剂量组),连续30天。观察大鼠体质量、前列腺湿重、前列腺指数的变化及前列腺组织病理学表现。结果:30天后与空白对照组比较,苯甲酸雌二醇高、中、低剂量组大鼠体质量降低(P<0.05);苯甲酸雌二醇中剂量组前列腺湿重升高(P<0.05),高、低剂量组前列腺湿重无明显差异(P>0.05);苯甲酸雌二醇中剂量组前列腺指数差异明显(P<0.05),高、低剂量组前列腺指数无明显差异(P>0.05);病理组织观察苯甲酸雌二醇中剂量组大鼠前列腺炎症较重,高、低剂量组无差异(P>0.05)。结论:连续多点皮下注射苯甲酸雌二醇能诱导去势大鼠慢性前列腺炎的产生且有一定的量效关系。
Objective:To establish experimental animal models with chronic nonbacterial prostatitis(CNP).Methods:Forty male SD rats were randomized into four groups,except blank control group,other three groups performed castrated operation after intraperitoneal anesthesia respectively,and accepted multi-point hypodermic injection of estradiol benzoate since the second day after the surgery,0.5,0.3 and 0.25 mg/kg(high,moderate and low dose groups),for 30 days consecutively.To observe body weight,wet weight of the prostate,prostate indexes changes and histopathological performance of the prostate of the rats.Results:Body weight of the rats in high,moderate and low doses groups of estradiol benzoate lowered when they were compared with blank control group in 30 days(P<0.05);wet weight of the prostate in moderate dose group of estradiol benzoate rose(P<0.05),no obvious difference could be found in wet weight of the prostate between high and low moderate dose groups(P>0.05);the difference was significant in prostate indexes in moderate dose group(P<0.05),the difference was insignificant in prostate indexes between high and low doses groups(P>0.05);pathological observation showed that inflammation of moderate dose group was more severe,no difference could be found between high and moderate doses groups(P>0.05).Conclusion:Continuous multi-point hypodermic injection of estradiol benzoate could induce chronic nonbacterial prostatitis in castrated rats,and it shows certain dose-effect relationship.
