摘要 自噬是一种重要的细胞再循环机制,负责降解细胞内不必要或功能失调的蛋白质和细胞器。体内有多条通路可以调节自噬过程,其中最重要的是哺乳动物雷帕霉素信号通路。自噬在代谢应激期间尤其活跃,在癌细胞中具有抑制肿瘤和促进肿瘤的双重作用,功能性自噬有助于预防坏死和炎症,导致肿瘤遗传基因不稳定;另一方面,在不利的代谢环境中,通过循环机制提供能量,自噬在肿瘤发生、发展的过程中扮演重要角色。临床治疗中关于诱导自噬还是抑制自噬对癌症治疗更有益处仍存在争论,针对自噬调节通路及过程研发的靶向药物在癌症治疗中作用显著。 Autophagy is an important cell recycling mechanism responsible for the degradation of unwanted or dysfunctional proteins and organelles in cells.There are multiple pathways in the body that regulate the autophagy process,the most important of which is the mammalian rapamycin signaling pathway.Autophagy is particularly active during metabolic stress.In cancer cells,it has the dual function of inhibiting tumors and promoting tumors.Functional autophagy helps prevent necrosis and inflammation,leading to instability of tumor genes;in an unfavorable metabolic environment,energy is provided through a circulatory mechanism,and autophagy plays an important role in the development of tumors.There is still debate in clinical about whether induction of autophagy or inhibition of autophagy is better for cancer treatment,and targeted drugs developed for autophagy regulatory pathways and processes play a significant role in the treatment of cancer.
