微小RNA-34a对乳腺癌细胞增殖和侵袭的抑制作用 被引量:1

摘要 目的:检测乳腺癌细胞中miR-34a的相对表达水平及其对细胞增殖和侵袭的影响。方法:采用qRT-PCR检测乳腺癌细胞MCF-7和乳腺正常细胞MCF-10A中miR-34a的相对表达水平;miR-34amimic(miR-34a模拟物,对照质粒)和miR-34a质粒转染至MCF-7细胞中,MTT实验检测MCF-7、MCF-7-mimic和MCF-7-miR-34a细胞的增殖情况;Transwell实验检测MCF-7、MCF-7-mimic和MCF-7-miR-34a细胞的侵袭能力;Western blot实验检测MCF-7、MCF-7-mimic和MCF-7-miR-34a细胞中Notch1蛋白表达。结果:乳腺癌细胞中miR-34a的表达水平低于乳腺正常细胞(P<0.05);转染实验使MCF-7-miR-34a组miR-34a的相对表达水平明显高于MCF-7和MCF-7-mimic组(P<0.001);MTT实验中60和72 h时MCF-7-miR-34a组细胞的吸光度值明显低于MCF-7和MCF-7-mimic组(P<0.05);MCF-7-miR-34a组发生侵袭的细胞数明显少于MCF-7和MCF-7-mimic组(P<0.05);MCF-7-miR-34a组与MCF-7组和MCF-7-mimic组相比Notch1蛋白的表达明显较弱。结论:miR-34a在乳腺癌细胞中表达水平降低,其可通过减弱Notch1蛋白的表达抑制乳腺癌细胞的增殖和侵袭。 Objective:To detect the relative expression of miR-34a in breast cancer cells and its effect on cell proliferation and invasion.Methods:Relative expression levels of miR-34a in breast cancer cell MCF-7 and normal breast cell MCF-10A cells were detected by qRT-PCR.;miR-34a-mimic and miR-34a plasmids were transfected into MCF-7 cells;MTT assay for prolifera-tion of MCF-7,MCF-7-mimic and MCF-7-miR-34a cells;Transwell assay for invasive ability of MCF-7,MCF-7-mimic and MCF-7-miR-34a cells,Western blot analysis of Notch1 protein expression in MCF-7,MCF-7-mimic and MCF-7-miR-34a cells.Results:The expression level of miR-34a in breast cancer cells was lower than that in normal breast cells(P<0.05);The relative expression level of miR-34a in MCF-7-miR-34a group was significantly higher than that in MCF-7 and MCF-7-mimic group(P<0.001);The A value of MCF-7-miR-34a cells at 60 and 72 h in MTT assay was significantly lower than that of MCF-7 and MCF-7-mimic groups(P<0.05);The number of cells invading in MCF-7-miR-34a group was significantly lower than that in MCF-7 and MCF-7-mimic group(P<0.05);The expression of Notch1 protein was significantly weaker in MCF-7-miR-34a group compared with MCF-7 and MCF-7-mimic groups.Conclusion:The expres-sion level of miR-34a is decreased in breast cancer cells.miR-34a inhibits proliferation and invasion of breast cancer cells by attenuating the expression of Notch1 protein.
