摘要
建立以芽为试材的软枣猕猴桃DNA提取方法,可实现样品的随时采集和ISSR分析。以软枣猕猴桃品种“桓优1号”为试材,分别采集展叶期的叶片和落叶期的芽,用改进的CTAB法提取基因组DNA并进行ISSR检测。结果表明:两种试材提取的DNA无降解,OD260/OD280值介于1.72~1.85,纯度和完整性均较好,两者ISSR扩增条带一致。该方法为后续软枣猕猴桃种质资源的鉴定和评价提供了技术保障。
The purpose of this study was to establish a DNA extracting method from buds of Actinidia arguta,which could collect samples and do ISSR analysis easily.In this experiment,taking“Huanyou No.1”as test material,leaves in leaf development stage and buds in deciduous stage were collected,genomic DNA were extracted by modified CTAB method and ISSR analyses were conducted.The result indicated that DNA extracted from the two samples were not degraded with OD260/OD280 values between 1.72 and 1.85,purity and integrity of DNA were good,and the ISSR amplification bands of the two samples were consistent.This method provided technical support for the identification and evaluation of A.arguta germplasm resources.
