摘要
目的探讨高效、简便、经济体外培养人成纤维细胞模型的建立培养方式。方法选择一位伤后5个月的5岁骨科患者,取得本人及其家属知情同意,证实术前未进行任何药物治疗,取其伤后红色的稍高出周边皮肤的瘢痕处小块组织,将组织分为A组和B组,其中A组采用组织块培养法,B组在组织块培养法的基础上使用0.25%胰蛋白酶和0.02%依地酸4 oC消化的改进组织块培养法。比较两种方法获得细胞的培养时间,细胞的生长情况和形态以及人成纤维细胞百分比。结果B组获得细胞的培养时间明显短于A组(P<0.05)。B组细胞的组织对比A组而言,贴壁更早更紧且不易漂浮,细胞核更大更明显。B组的细胞百分比和干细胞含量均明显多于A组(P<0.05)。结论采用改进组织块培养法体外培养人成纤维细胞模式简单高效,能够比常规组织块更快地培育细胞,对细胞生物学的研究意义重大。
Objective To study the establishment of human fibroblast model in vitro with high efficiency,convenience and economy.Methods I chose a 5-year-old orthopedic patient who was 5 months after the injury and obtained informed consent from the person and her family.It was confirmed that no medical treatment was performed before the operation,and the small red tissue slightly above the scar of the surrounding skin was taken after the injury.The tissues were divided into group A and group B.Group A was treated with tissue block culture method,and group B was modified tissue block culture method using 0.25%trypsin and 0.02%edetate 4 oC digestion on the basis of tissue block culture method.The two methods were compared to obtain cell culture time,cell growth and morphology,and percentage of human fibroblasts.Results Cell culture time of group B was significantly shorter than the group A(P<0.05).Compared with group A,the tissues of cells in group B adhered to the wall earlier,tighter and less likely to float,and the nuclei were larger and more obvious.Cell percentage and stem cell content in group B were significantly higher than the group A(P<0.05).Conclusion It is simple and efficient to cultivate human fibroblasts in vitro by improved tissue mass culture method,which can grow cells faster than conventional tissue mass,and is of great significance to the study of cell biology.
