摘要
目的探讨蛋白激酶Cα(PKCα)与Polo样激酶1(PLK1)在膀胱癌细胞中的表达及其相关性。方法利用PKCα的siRNA转染膀胱肿瘤J82细胞系后,基因芯片筛选出下游靶点PLK1,RT-PCR及Western-blot方法对其表达水平进行验证,分析两者的相关性。结果在膀胱癌细胞中利用siRNA抑制PKCα的表达会导致膀胱癌细胞中PLK1的表达量的下降(P<0.05)。同时在膀胱肿瘤标本中二者呈正相关(P<0.01)。结论 PKCα和PLK1是膀胱癌治疗的可能新靶点。
Objective To investigate the expression and correlation of Protein Kinase Cα(PKCα) and Pololike Kinase 1(PLK1) in bladder cancer J82 cells. Methods siRNA of PKCαwas transfected into the bladder tumor J82 cell line, the downstream target PLK1 was selected by gene chip. RT-PCR and Western-blot were used to verify the expression of PKCαand PLK1 and their correlation was analyzed. Results The inhibition of PKCα expression down-regulated the expression level of PLK1(P<0.05) in bladder tumor J82 cells. The expression of PKCα was positively related to PLK1 expression in bladder cancer J82 cells(P<0.01). Conclusion PKCα and PLK1 are possible new target for bladder cancer treatment.
