五味子醇提液对糖尿病肾病小鼠氧化应激的保护作用及机制研究 被引量:28

Protective effect and mechanism of Schisandra chinensis extract on oxidative stress in diabetic nephropathy mice

摘要 目的探讨五味子醇提液(SE)对自发型2型糖尿病C57BLKS/Jdb/db(db/db)小鼠氧化应激的保护作用及机制。方法选取10只9周龄雄性db/m小鼠和10只同周龄同源雄性db/db小鼠随机分为对照组、SE对照组、模型组、SE治疗组,每组5只。SE对照组和治疗组分别以SE 5 mg/kg ig给药,连续给药6周。在0、3、6周记录体质量、血糖及24 h尿微量白蛋白,6周后处死小鼠,收集肾组织标本。脂质过氧化试剂盒测定氧化应激指标丙二醛(MDA)含量,过碘酸雪夫(PAS)染色观察肾组织病理变化,Western blotting法检测抗氧化因子核因子红细胞衍生2相关因子2(Nrf2)及血红素氧合酶-1(HO-1)蛋白表达,实时荧光定量PCR(qRT-PCR)法检测Nrf2及其下游靶基因的表达。结果与对照组相比,模型组小鼠体质量、血糖和24 h尿微量白蛋白明显升高,MDA含量增加,Nrf2、HO-1及其下游靶基因表达下降(P<0.05、0.01)。SE能够降低糖尿病肾病小鼠血糖、24 h尿微量白蛋白和MDA水平,改善肾组织病理损伤,上调Nrf2、HO-1及其下游靶基因表达(P<0.05、0.01)。结论SE对db/db小鼠氧化应激损伤有一定的保护作用,其机制可能与上调抗氧化因子Nrf2及其下游基因有关。 Objective To investigate the protective effect of Schisandra chinensis extract on oxidative stress in db/db mice,and to explore its possible mechanism.Methods Ten male 9-week-old db/m mice and ten homologous male db/db mice of the same age were randomly divided into normal control group(C+V group),S.chinensis extract control group(C+SE group),model group(DN+V group),and S.chinensis extract-treated group(DN+SE group),with five mice in each group.The S.chinensis extract control group and S.chinensis extract-treated group were respectively ig administrated with 5 mg/(kg·d)of S.chinensis extract for 6 weeks.The body weight,blood glucose and 24 h urine micro-albumin were recorded at 0,3,and 6 weeks.After 6 weeks,the mice were sacrificed and their kidney tissue specimens were collected.The oxidative stress index of malondialdehyde(MDA)were examined by lipid peroxidation kit.The pathological changes of kidney tissues were observed by PAS staining.The expression of antioxidant factor Nrf2 and HO-1 protein of kidney tissue was detected by Western Blotting.The expression of Nrf2 and its downstream target genes of kidney tissue were detected by qRT-PCR.Results Compared with the normal control group,the body weight,blood glucose and 24h urine micro-albumin of the model group were significantly increased and the MDA content were increased.Therefore,the renal tissue pathological damage aggravated,and the expression of Nrf2 and HO-1,as well as its downstream target genes were all downregulated.(P<0.01 or P<0.05).S.chinensis extract reduced blood glucose,24 h urine micro-albumin and MDA level in diabetic nephropathy mice,improved renal pathological damage,and up-regulated Nrf2 and HO-1 and its downstream target gene expression.(P<0.01 or P<0.05).Conclusion S.chinensis extract has protective effect on oxidative stress injury in db/db mice,and its mechanism may be related to up-regulation of antioxidant factor Nrf2 and its downstream genes.
