摘要
蔗糖转化酶SUC2是酿酒酵母水解菊糖的关键酶,但SUC2在酿酒酵母中的低水平表达限制了其理性改造酿酒酵母以获得高效生产菊糖基乙醇工程菌的进程。本实验通过克隆获得蔗糖转化酶SUC2的编码基因,并利用AGA1-AGA2锚定蛋白复合体将SUC2表面展示于商品化酿酒酵母菌株EBY100中。酶活力测定结果表明,表面展示有SUC2的重组菌EBY-S蔗糖酶活及菊糖酶活分别达到241.6和9.4 U/mL,较之前报道的过表达SUC2的酿酒酵母重组菌酶活力有显著提高。该结果表明,AGA1-AGA2锚定蛋白复合体能够成功地将SUC2表面展示于酿酒酵母细胞壁上。
The sucrose invertase SUC2 is the key enzyme in the hydrolysis of inulin by Saccharomyces cerevisiae, but the low expression level of SUC2 in Saccharomyces cerevisiae limits the process of rational engineering of Saccharomyces cerevisiae to obtain an efficient inulin based ethanol producing strain. The gene encoding SUC2 was cloned and displayed on the surface of commercial Saccharomyces cerevisiae strain EBY100 using AGA1-AGA2 anchoring protein complex. The results showed that the invertase and inulinase activities of SUC2 on the surface of EBY-S were 241.6 and 9.4 U/mL, respectively, compared with the previously reported Saccharomyces cerevisiae recombinant overexpressed SUC2. The results indicated that the surface of SUC2 could be successfully displayed on the cell wall of Saccharomyces cerevisiae by AGA1-AGA2 anchoring protein complex.
