不同甘薯品种及其辐射诱变后代基因组变异的分子标记评价 被引量:2

Molecular Marker Evaluation of Genomic Variation of Different Sweetpotato Varieties and Their Radiation Induced Progenies

摘要 利用SCoT、ISSR、CBDP等3种分子标记技术,对2个优质甘薯品种川薯34和川紫薯2号的60Co-γ辐射诱变后代进行基因组变异分析。结果表明,6条SCoT引物在参试材料中最多可扩增出40条条带,平均每条引物扩增6.67条,其中,在川薯34诱变后代中共检测到变异条带4条,在川紫薯2号诱变后代中共检测到变异条带7条;2条ISSR引物最多可扩增出6条条带,平均每条引物扩增3.0条条带,在川薯34的后代中发现变异条带1条,在川紫薯2号的后代中没有发现变异条带;6条CBDP引物最多可扩增出36条条带,平均每条引物可扩增6条条带,在川薯34和川紫薯2号的后代中分别发现变异条带13条和7条。遗传相似性分析表明,SCoT、ISSR和CBDP等3种分子标记在川薯34诱变后代中的遗传相似性变异范围为0.80~1.00;在川紫薯2号诱变后代的遗传相似性系数变异范围为0.86~1.00。研究结果显示,SCoT分子标记和CBDP分子标记的扫描结果均优于ISSR分子标记,且CBDP分子标记结果最佳。聚类分析结果表明,川紫薯2号和川薯34诱变后代的部分单株未发生变异,部分单株间存在变异,个别单株间遗传相似性差异较大。研究结果初步证明,悬浮细胞低剂量照射产生的甘薯诱变后代个体间变异较小,且突变程度与品种相关。研究结果将为分子标记技术和核辐射诱变育种技术在甘薯育种中的应用提供参考。 This study used SCoT,ISSR and CBDP three kind of molecular markers to analyze the genomic variation of 60Co-γradiation mutagenized offspring of two sweetpotato varieties Chuanshu 34 and Chuanzishu 2.The results showed that a total of 40 bands were amplified by the six SCoT primers in the test materials,and an average of 6.67 products was amplified.4 variant bands were detected in the progeny of Chuanshu 34,7 variant bands were detected in the progeny of Chuanzishu 2.2 ISSR primers amplified 6 bands,and each of the primers amplified 3.0 bands.One variant band was found in the offspring of Chuanshu 34,and no variant band was found in the offspring of Chuanzishu 2.36 bands were amplified by 6 CBDP primers,and each primer amplified the 6.0 bands.In the offspring Chuanshu 34 and Chuanzishu 2,13 and 7 variant bands were found,respectively.Genetic similarity analysis showed that the genetic similarity variation of three molecular markers of SCoT,ISSR and CBDP was 0.80-1.00 in the mutants of Chuanshu 34,the variation of genetic similarity coefficient was 0.86-1.00 in the mutants of Chuanzishu 2.The results indicated that CBDP marker was better than SCoT and ISSR markers in scanning of sweetpotato genome mutant.The results of cluster analysis indicated that there were small variations among many mutants of Chuanzishu 2 and Chuanshu 34,and some larger variations were found in few mutants.The results preliminarily proved that the mutation of the progeny of sweetpotato induced by low dose irradiation of suspension cells was relatively small,and the degree of mutation was related to the variety.These results will provide reference for the application of molecular marker and nuclear radiation mutation breeding in sweetpotato breeding.
