摘要
为了比较不同产地桃叶中多糖含量及其体外抗氧化活性的差异,选取4个不同产地桃叶,采用水提醇沉法制备多糖,以紫外分光光度计法测定多糖得率,同时通过体外清除DPPH·、ABTS^+·、·OH、O2^-·自由基的方法,测定桃叶多糖的抗氧化活性。结果表明:各产地桃叶的多糖得率由高到低排列依次为山东沂水(2.49±0.05)%>浙江奉化(2.25±0.04)%>湖北随州(2.06±0.03)%>湖南衡阳(1.78±0.04)%,不同产地桃叶多糖对DPPH·、ABTS^+·、·OH、O2^-·自由基的半数抑制浓度IC50值均表现为湖南衡阳>湖北随州>浙江奉化>山东沂水,故其抗氧化活性由强到弱依次为山东沂水>浙江奉化>湖北随州>湖南衡阳。这表明,不同产地桃叶中总多糖含量存在较大差异,且抗氧化活性与多糖含量具有正相关性。
To compare the differences of polysaccharide contents in peach leaves and of their antioxidant activity in vitro,peach leaves from four producing areas were extracted by heating reflux with water as solvent,and the polysaccharide content was determined by ultraviolet spectrophotometer.Meanwhile,the antioxidant activity of polysaccharide from peach leaf was determined by scavenging DPPH·,ABTS^+·,·OH,O2^-·free radicals in vitro.The results showed that the polysaccharide contents had a big difference between peach leaves of different areas,which from high to low was Yishui,Shandong(2.49±0.05%)>Fenghua,Zhejiang(2.25±0.04%)>Suizhou,Hubei(2.06±0.03%)>Hengyang,Hunan(1.78±0.04%),the IC50 values of DPPH·,ABTS^+·,·OH and O2^-·free radicals were Hengyang,Hunan>Suizhou,Hubei>Fenghua,Zhejiang>Yishui,Shandong,and the antioxidant activity was Yishui,Shandong>Fenghua,Zhejiang>Suizhou,Hubei>Hengyang,Hunan.There is a positive correlation between antioxidant activity and polysaccharide content.
