摘要
为探讨小麦蔗糖转运蛋白5 (sucrose transporter 5,SUT5)蛋白的数量、结构特征和性质差异,以水稻SUT为模板,BlastP获得21条小麦SUT蛋白序列,其中六条鉴定为小麦SUT5蛋白。将其和水稻SUT5蛋白一起,进行了蛋白质亚细胞定位、基序、二级结构、跨膜结构以及磷酸化位点等相关分析。发现六条小麦SUT5蛋白和水稻SUT5都含有MFS_2基序,但水稻SUT5含有BT_1基序,小麦SUT5含有PUCC基序。小麦和水稻的SUT5在跨膜结构域处的二级结构较为一致,但N端和C端差异较大,且仅有SUT5-4与水稻SUT5的跨膜结构域数量一致。磷酸化位点预测结果显示,小麦和水稻的SUT5蛋白在C末端都存在丝氨酸的磷酸化位点,N末端差异较大。本研究为小麦SUT5蛋白亚家族的研究和利用提供了理论参考及分析依据。
To investigate the number,structure,characteristics of sucrose transporter 5(SUT5)proteins in wheat,21 wheat SUT proteins were got by BlastP program taking rice SUT as the template.6 proteins were identified as wheat SUT5s.Coupled with OsSUT5,wheat SUT5 proteins were analyzed to subcellular location,motif,secondary structure,transmembrane domain,and phosphate sites.The results showed that MFS_2 motif were found in all SUT5 proteins,but BT_1 motif was found in rice SUT5 only.PUCC motif was found in wheat SUT5 only.The secondary structure of SUT5 in rice and wheat were similar in transmembrane domain.The N-and C-terminus were quite different.Moreover,only SUT5-4 has the same number of transmembrane domain as rice SUT5.Similar phosphate site(Ser)was predicted in C-terminus of all SUT5 proteins,not in N-terminus.This research might provide help for the follow-up study of wheat SUT5 subfamily.
