槲寄生联合RIPOC对大鼠心肌保护作用 被引量:2

摘要 目的:探讨槲寄生联合远隔缺血后适应(RIPOC)对心梗大鼠心肌的保护作用。方法:56只健康成年SD大鼠按随机数字表法分为模型组(n=10)、槲寄生组(n=12)、RIPOC组(n=12)、联合组(n=12)及对照组(n=10)。模型组、槲寄生组、RIPOC组及联合组均建立急性心肌梗死动物模型,对照组只进行开胸。槲寄生组给予槲寄生有效提取物;RIPOC组进行RIPOC操作;联合组给予槲寄生有效提取物+RIPOC操作;对照组、模型组与RIPOC组均给予等量0.9%氯化钠溶液,均持续干预14 d。比较各组心功能指标、左室质量指数(LVMI)、左心室壁厚度、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)含量。结果:模型组左心室射血分数(LVEF)、SOD及GSH-Px均明显低于对照组,而左心室舒张末期内径(LVEDV)、左心室收缩末期内径(LVESV)、LVMI、左心室壁厚度及MDA均高于对照组,差异均有统计学意义(P<0.05)。槲寄生组、RIPOC组和联合组LVEF、SOD及GSH-Px均高于模型组(P<0.05);槲寄生组、RIPOC组和联合组LVEDV、LVESV、LVMI、左心室壁厚度、MDA均低于模型组(P<0.05);联合组GSH-Px和SOD水平均高于槲寄生组和RIPOC组,而MDA均低于槲寄生组和RIPOC组,差异均有统计学意义(P<0.05)。结论:槲寄生、RIPOC均可改善心梗大鼠心功能,逆转心肌肥厚和改善心肌氧化应激反应,且槲寄生联合RIPOC改善心肌氧化应激反应效果更加显著,两者可以协同保护缺血性心脏,值得推广。 Objective:To investigate the protective effect of Viscum Coloratum combined with remote ischemic postconditioning(RIPOC)on myocardium of myocardial infarction rats.Method:A total of 56 healthy adult SD rats were divided into model group(n=10),Viscum Coloratum group(n=12),RIPOC group(n=12),combination group(n=12)and control group(n=10)according to the random number table method.Animal models of acute myocardial infarction were established in the model group,Viscum Coloratum group,RIPOC group and combination group,while thoracotomy was performed only in the control group.The Viscum Coloratum group was treated with the effective extract of Viscum Coloratum.The RIPOC group was treated with RIPOC operation.The combination group was treated with the effective extract of Viscum Coloratum+RIPOC operation.The control group,the model group and the RIPOC group were given equivalent 0.9% Sodium Chloride Solution,and all of them continued to intervene for 14 days.The cardiac function index,left ventricular mass index(LVMI),left ventricular wall thickness,superoxide(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in all group were compared.Result:Left ventricular ejection fractions(LVEF),SOD and GSH-Px in the model group were significantly lower than those in the control group,while left ventricular end-diastolic diameter(LVEDV),left ventricular end-systolic diameter(LVESV),LVMI,left ventricular wall thickness and MDA were higher than those in the control group,the differences were statistically significant(P<0.05).LVEF,SOD and GSH-Px in the Viscum Coloratum group,RIPOC group and the combination group were all higher than those in the model group(P<0.05).LVEDV,LVESV,LVMI,left ventricular wall thickness and MDA in the Viscum Coloratum group,RIPOC group and the combination group were all lower than those in the model group(P<0.05).GSH-Px and SOD in the combination group were higher than those in the Viscum Coloratum group and the RIPOC group,while MDA in the combination group were lower than those in the Viscum Coloratum group and the RIPOC group(P<0.05).Conclusion:Viscum Coloratum and RIPOC can both improve cardiac function,reverse myocardial hypertrophy and improve myocardial oxidative stress response in rats with myocardial infarction,and the effect of Viscum Coloratum combined with RIPOC on myocardial oxidative stress response is more significant.They can coordinate to protect ischemic heart,which is worthy of promotion.
