摘要 〔目的〕通过探究NME基因家族成员表达水平和乳腺癌不同亚型、临床病理特征及预后的关系,以期阐明NME基因家族成员在乳腺癌中的潜在作用。〔方法〕利用Oncomine数据库.Kaplan-Meier plotter数据库、人类蛋白质图谱数据库(The Human Protein Atlas)分析乳腺癌NME所有家族成员的mRNA、蛋白质表达及其与乳腺癌患者整体生存率的相关性等内容。〔结果〕(1)NME1、NME2、NME3和NME4 mRNA在大多数癌症中显著高表达,而NME5则低表达。NME1、NME2、NME3、NME8、NME9和NME10在乳腺癌中高表达。(2)NME1、NME2低表达和NME3、NME5、NME7高表达乳腺癌患者的整体生存率较好。对于不同亚型乳腺癌患者,不同NME家族成员有不同的预后表现。(3)NME2高表达和NME3、NME5、NME9及NME8低表达分别与不同淋巴结状态和p53状态的乳腺癌患者整体生存率恶化有关。〔结论〕(1)NME1、NME2、NME3、NME5和NME7可以作为乳腺癌的预后生物标志物。(2)NME1、NME3、NME5、NME7和NME9可以作为不同乳腺癌亚型的预后生物标志物,其中NME1低表达和NME5 mRNA水平高表达是Luminal A型乳腺癌良好预后标志物,而NME3、NME5 mRNA水平低表达和NME9 mRNA水平高表达水平高表达对于Basal like型乳腺癌有较好的预后作用,NME7 mRNA水平高表达对于HER2+型乳腺癌有较好的预后作用。(3)NME家族成员在乳腺癌组织中的mRNA表达水平和患者预后、病理分级、淋巴结状态和P53状态相关。 [Objective]Exploring the relationship between the expression level of all members of NME family and the prognosis of breast cancer patients in different subtypes will promote elucidating the potential functional role of NME family members in breast cancer.[Methods]The mRNA and protein expression levels of all NME family members in breast cancer patients were examined by using Oncomine database and The Human Protein Atlas,and the correlation between NME and overall survival of breast cancer patients was analyzed by Kaplan-Meier plotter.[Results](1)The expression levels of NME1,NME2,NME3 and NME4 mRNA were significantly higher in most cancer tissues than that in normal tissues,while the expression level of NME5 was down-regulated.The mRNA levels of NME1,NME2,NME3,NME&NME9 and NME10 were up-regulated in breast cancer.(2)Breast cancer patients with the lower expression of NME1 and NME2 and the higher expression of NME3,NME5 and NME7 showed better prognosis.For different subtypes of breast cancer patients,different NME family members have different manifestations.(3)The higher expression of NME2 and the lower expression of NME5 and NME9 and NME8 were associated with over survival(OS)deterioration in breast cancer patients with different lymph node status and p53 status,respectively.〔Conclusion〕(1)NME1,NME2,NME3,NME5 and NME7 can be used as prognostic biomarkers for all breast cancer.(2)NME1,NME3,NME5,NME7 and NME9 can be as different subtypes of breast cancer prognosis biomarkers,and lower NME1 and higher NME5 expression for the prognosis of Luminal A breast cancer are the good markers.Lower NME3,NME5 and higher NME9 expression all can predict better prognosis for Basal like breast cancer patients,higher NME7 expression predicting better prognosis for HER2+breast cancer.(3)The mRNA expression level and prognostic value of NME family in breast cancer patients were significantly correlated with pathological grade,lymph node status and p53 status.
