摘要
为研究虎杖苷(PD)对热应激大鼠模型空肠消化吸收、抗氧化和肠屏障功能的影响,采用2×2设计,将24只雄性SD大鼠随机分为4组(n=6):室温处理+灌胃羧甲基纤维素钠(CMC-Na)溶液组、室温处理+灌胃PD溶液组、热应激处理+灌胃CMC-Na溶液组、热应激处理+灌胃PD溶液组。热应激组每天40℃处理1.5 h,连续3 d。结果显示,在热处理后大鼠空肠绒毛高度、绒毛高度隐窝深度比值、蔗糖酶和麦芽糖酶活性下调。热处理还降低了空肠GSH含量和T-AOC活力;PD处理提高了GSH含量和SOD基因表达量,降低了GSH-PX活力。热处理降低了CLDN2基因表达量;PD处理增加了CLDN2、CLDN3、ZO-1基因表达量。热处理提高了空肠组织TLR4、NF-κB、TNF-α和SIRT1基因表达量及细胞因子IL-6、IL-10、TNF-α含量;PD处理提高了SIRT1基因表达量,降低了IL-10含量。结果表明,虎杖苷具有改善热应激大鼠肠道氧化应激和屏障损伤的积极作用。
To study the effect of polydatin(PD) on jejunal digestion,absorption,anti-oxidation and intestinal barrier function in a heat stress rat model,24 male SD rats were randomly divided into 4 groups using 2×2 designs(n=6):room temperature treatment+gavage sodium carboxymethyl cellulose by intragastric administration(CMC-Na) solution,room temperature treatment+gavage PD solution,heat stress treatment+gavage CMC-Na solution,heat stress treatment+gavage PD solution.The heat stress groups were treated at 40 ℃ for 1.5 h/d for 3 consecutive days.The results showed that the measurements were down-regulated after heat treatment including rat intestinal villus height,villus to crypt ratio,sucrase and maltase activity.Heat treatment also reduced GSH content and T-AOC activity in jejunum;PD treatment increased GSH content,SOD gene expression and decreased GSH-PX activity.Heat treatment reduced the expression of CLDN2 gene;PD treatment raised the expression of CLDN2,CLDN3 and ZO-1 genes.Heat treatment increased TLR4,NF-κB,TNF-α and SIRT1 gene expression and cytokines IL-6,IL-10 and TNF-α levels in jejunum tissues;PD treatment increased SIRT1 gene expression and decreased IL-10 content.In conclusion, the addition of polydatin has a positive effect on improving intestinal oxidative stress and barrier damage in rats with heat stress.
