不同浓度地塞米松对慢性哮喘大鼠肺部组织学干预作用 被引量:3

摘要 目的观察不同浓度地塞米松对慢性哮喘大鼠一般情况及肺部组织学的干预作用,探索地塞米松(地米)在慢性哮喘大鼠模型中适合的灌胃剂量。方法75只雄性SD大鼠,应用随机数字表法分为正常组、模型组及地米低剂量、中剂量和高剂量组。卵清蛋白致敏和激发法构建慢性哮喘大鼠模型。从实验第20天,地塞米松各组分别按0.0625、0.125、0.5 mg·kg^-1·d^-1灌胃给药。观察记录大鼠一般情况,HE染色法、Masson染色法观察大鼠肺部组织学改变。结果模型组大鼠支气管及周围组织炎性浸润程度较正常组增加,气道壁和平滑肌的厚度以及气道胶原的沉积均明显高于正常组(P<0.01)。地米高剂量组干预一周后大鼠日均进食量及体质量均明显低于模型组(P<0.01);地米中、低剂量组干预后各周体质量增加较模型组明显减缓(P<0.01);地米中、低剂量组大鼠日均进食量在干预2周后较模型组明显减少(P<0.01),地米中剂量组大鼠日均进食量在干预4周后明显少于低剂量组(P<0.01);大鼠肺组织炎性浸润情况地米中、低剂量组较模型组明显减轻,气道壁和平滑肌厚度以及气道胶原沉积亦明显降低(P<0.01),二组间无差异(P>0.05);地米高剂量组大鼠在干预5次后大量死亡。地米中剂量组干预第5周死亡3只,余组均无死亡。结论地塞米松中、低剂量均可用作慢性哮喘大鼠实验中的阳性对照剂量,但低剂量后期副作用更少,故地塞米松低剂量在慢性哮喘大鼠实验中可能是更安全、合适的灌胃剂量。 Objective To observe the intervention effects of different concentrations of dexamethasone on the general situation and pulmonary histology of chronic asthma rats,and to explore the suitable intragastric dose of dexamethasone in the rat model of chronic asthma.Methods Totally 75 healthy male Sprague-Dawley(SD)rats were randomly divided into normal group,model group,dexamethasone low dose,middle dose and high dose group by random number table.The rat model of chronic asthma was established by sensitization and challenge of ovalbumin(OVA).From the 20th day of the experiment,dexamethasone groups were given intragastric administration of 0.0625 mg/kg/d,0.125 mg/kg/d and 0.5 mg/kg/d,respectively.The general situation of rats was observed and recorded.The histological changes of rat lungs were observed by HE staining and Masson staining.Results The degree of inflammatory infiltration of bronchus and surrounding tissue in the model group was significantly higher than that in the normal group(P<0.01).The thickness of airway wall and smooth muscle and the deposition of airway collagen in the model group were significantly higher than those in the normal group(P<0.01).After one week of intervention,the average daily food intake and body mass of rats in the high dose group were significantly lower than those in the model group(P<0.01),and the increase in body mass in the middle and low dose groups was significantly slower than that in the model group(P<0.01).After 2 weeks of intervention,the average daily food intake of rats in the middle and low dose groups was significantly lower than that in the model group(P<0.01).After 4 weeks of intervention,the average daily food intake in the middle and low dose groups was significantly lower than that in the low dose group(P<0.01).The inflammatory infiltration of lung tissue in the middle and low dose groups was significantly less than that in the model group,and the thickness of airway wall and smooth muscle and the deposition of airway collagen in the middle and low dose groups were significantly lower than those in the model group(P<0.01).There was no difference between the two groups(P>0.05).A large number of rats in the high dose group died after 5 times of intervention.Three rats died in the middle dose group at the 5th week of intervention,and there was no death in the other group.Conclusions Both middle and low doses of dexamethasone can be used in the rat model of chronic asthma,but the late side effects of low dose of dexamethasone are less,so the low dose of dexamethasone may be more suitable for the rat model of chronic asthma.
