电针辅助治疗对阿尔茨海默病大鼠认知功能和学习记忆能力的影响分析 被引量:5

摘要 目的分析电针辅助治疗对阿尔茨海默病(AD)大鼠认知功能、学习记忆能力的影响,为AD的临床干预提供实验数据。方法60只清洁级雄性Wister大鼠60只清洁级雄性Wister大鼠按随机数字表法分为正常组、假手术组、模型组、西药组、电针辅治组,每组8只;模型组、西药组和电针辅治组双侧脑室注射β淀粉样蛋白25-35(Aβ25-35)诱导AD大鼠模型;西药组、电针辅治组采用电针治疗2周,电针频率分别为2 Hz、50 Hz;假手术组双侧脑室0.9%氯化钠注射,且正常组、模型组及假手术组仅在电针组大鼠行电刺激时给予同样的抓取及固定动作,不进行其他处理;采用Morris水迷宫测试行定位航行实验、空间探索实验并Morris水迷宫测试结束后取大鼠海马及皮质组织;采用免疫印迹检测法(WB)检测大鼠海马区及皮质组织的糖原合酶激酶(GSK-3β)蛋白表达及GSK-3β第9位的丝氨酸(Ser-9)和216位的酪氨酸(Try-216)水平。结果①模型组各时间点的逃避潜伏期、首次跨越时间明显较正常组长,跨越平台次数明显较正常组少提示造模成功;而较模型组,西药组、电针辅治组各个时间点的逃避潜伏期、首次跨越时间明显缩短跨越平台次数明显增多,且电针辅治组的各个时间点的逃避潜伏期、次跨越平台时间明显短于西药组,跨越平台次数明显较西药组多差异均有统计学意义(P<0.05);②较正常组及假手术组,模型组海马及皮质组织GSK-3β、GSK-3β(Try-216)明显升高,GSK-3β(Ser-9)明显降低;而与模型组比较,西药组及电针辅治组海马及皮质组织GSK-3β、GSK-3β(Try-216)均下降,GSK-3β(Ser-9)则上升,且电针辅治组海马及皮质组织GSK-3β、GSK-3β(Try-216)显著低于西药组,GSK-3β(Ser-9)显著高于西药组,差异有统计学意义(P<0.05)。结论电针辅助治疗对阿尔茨海默病大鼠认知功能、学习记忆能力的改善优于西药干预。 Objective To analyze effects of electroacupuncture adjuvant therapy on cognition function,learning and memory ability in rats with Alzheimers disease(AD),and to provide experimental data for AD clinical intervention.Methods Sixty clean-grade male Wister rats were divided into normal group,sham operation group,model group,western medicine group and electroacupuncture adjuvant therapy group,8 rats in each group.The bilateral ventricular injection of amyloid β-protein 25-35(Aβ25-35) was conducted to induce AD rat models.Western medicine group and electroacupuncture adjuvant therapy group were treated with electroacupuncture for 2 weeks,with electroacupuncture frequency of 2 Hz and 50 Hz.The sham operation group was injected with 0.9% sodium chloride in bilateral ventricle.The normal group,model group and sham operation group were given the same grasping and fixation action only when the rats in electroacupuncture adjuvant therapy group were given electrical stimulation,and they were not given other treatment.The Morris water maze test was applied in place navigation test and space exploration experiment.After Morris water maze test,rats hippocampus and cortical tissues were collected.Western Blotting(WB) was applied to detect expression of Glycogen synthase kinase-3(GSK-3β) protein and levels of serine(Ser-9) at 9 th site and tyrosine(Try-216)at position 216 th site of GSK-3β in hippocampus and cortex tissues of rats.Results①The escape latency and the first spanning time in model group were significantly longer than those in normal group,while number of spanning platforms was significantly less than that in normal group,suggesting successful modeling.Compared with model group,escape latency and first spanning time were significantly shortened in western medicine group and electroacupuncture adjuvant therapy group,while number of spanning platforms was significantly increased.The escape latency and first platforms spanning time in electroacupuncture adjuvant therapy group were significantly shorter than those in western medicine group,while number of spanning platforms was significantly more than that in western medicine group(P<0.05).②Compared with normal group and sham operation group,GSK-3β and GSK-3β(Try-216) in hippocampus and cortex tissues were significantly increased in model group,while GSK-3β(Ser-9) was significantly decreased.Compared with model group,GSK-3β and GSK-3β(Try-216) in hippocampus and cortex tissues were decreased in western medicine group and electroacupuncture adjuvant therapy group,while GSK-3β(Ser-9) was increased.The GSK-3β and GSK-3β(Try-216) in hippocampus and cortex tissues in electroacupuncture adjuvant therapy group were significantly lower than those in western medicine group,while GSK-3β(Ser-9) was significantly higher than that in western medicine group(P <0.05).Conclusion Electroacupuncture adjuvant therapy is better than western medicine in improving cognition function,learning and memory ability in AD rats.
