摘要
探究不同剂量的小分子金枪鱼多肽对小鼠记忆力、皮肤弹性和睡眠质量的影响。将6周龄的昆明小鼠随机分成4组(n=12),剂量组分别灌胃0.25 g/kg bw、0.5 g/kg bw和1.0 g/kg bw的小分子金枪鱼多肽,对照组灌胃PBS,连续6周。采用分光光度法分别测定小鼠脑组织SOD、GSH-Px和MDA含量,采用HE染色法制作小鼠皮肤石蜡切片,采用实时荧光定量PCR检测与记忆力、皮肤弹性和睡眠相关的基因表达。研究结果显示,小分子金枪鱼多肽能通过增加小鼠脑组织SOD和GSH-Px的含量,降低MDA的含量,上调记忆力相关基因的表达量来增强小鼠的记忆力;通过改善小鼠皮肤胶原纤维形态数目,改变皮肤弹性相关基因表达量来增加小鼠皮肤弹性;通过改变睡眠相关基因表达量来改善小鼠睡眠质量,且主要作用于GABAA通路和Hypocretin通路来发挥促眠作用。
It is explored that the effects of different doses of small molecule polypeptide of tuna on memory,skin elasticity and sleep quality in mice.There were random 4 groups,and each group included 126-week-old mice,small molecular tuna polypeptides of 0.25 g/kg bw,0.5 g/kg bw and 1.0 g/kg bw in the dose group,one of groups was continuously gavage PBS for 6 weeks,the others were continuously gavage different doses of small molecule polypeptide of tuna,respectively.Spectrophotometry was used to detect the contents of SOD,GSH-Px and MDA in brain of each group,respectively.HE staining was used to make paraffin sections of mice skin.And the expression levels of related genes of memory,skin elasticityand sleep were detected by qPCR.The research showed that the small molecule polypeptide of tuna could enhance the memory of mice by increasing the contents of SOD and GSH-Px,decreasing the contents of MDA in brain,and increasing the memory-related genes’expression levels.In addition,it can increase the skin elasticity of mice by improving the morphology of collagen fibers in skin and changing the expression levels of skin elasticity-related genes.Moreover,it can improve the sleep quality of mice by altering the expression levels of sleep-related genes,the GABAA pathway and Hypocretin pathway were mainly used to promote sleep.
