摘要
本文探索了西红花中西红花苷-Ⅰ和苷-Ⅱ的静态变化。利用色谱柱依利特hypersil ODS2(4.6 mm×250 mm,5μm),流动相为甲醇-水(45:55),流速0.8 mL/min,柱温为30℃,检测波长440 nm。结果表明,西红花苷-Ⅰ在1.207~120.36μg/mL(R^2=0.9998)范围内线性关系良好,平均回收率为97.13%(RSD=1.47%);西红花苷-Ⅱ在1.178~100.26μg/mL(R^2=0.9997)范围内线性关系良好,平均回收率为100.40%(RSD=1.39%)。西红花中西红花苷-1和西红花苷-Ⅱ的平均含量分别为10.07%和7.66%。固体样品和液体样品分别存放一年,西红花中西红花苷-Ⅰ和苷-Ⅱ的含量基本保持不变。该方法操作简便,结果准确。固体和液体样品中西红花苷-Ⅰ和苷-Ⅱ的含量在一年中的稳定性较好,可为西红花资源的进一步开发利用和质量评价提供基础。
This article explored the static changes of Crocin-Ⅰand Crocin-Ⅱin saffron.Using the column Elite hypersil ODS2(4.6 mm×250 mm,5μm),the mobile phase was methanol-water(45:55)at a flow rate of 0.8 mL/min,the column temperature was set at 30℃,and the detection wavelength was set at 440 nm.The results indicated that the Crocin-Ⅰhas a good linear relationship in the range of 1.207~120.36μg/mL(R^2=0.9998),with an average recovery of 97.13%(RSD=1.47%);the linear relationship of Crocin-Ⅱin the range of 1.178~100.26μg/mL(R^2=0.9997)was good,and the average recovery was 100.40%(RSD=1.39%).The average content of Crocin-1 and Crocin-Ⅱin saffron was 10.07%and 7.66%,respectively.The solid sample and the liquid sample were stored for one year respectively,and the contents of Crocin-Ⅰand Crocin-Ⅱof saffron remained basically unchanged.The method was simple and the results were accurate.The content of Crocin-Ⅰand Crocin-Ⅱin solid and liquid samples was stable in one year,which provided a basis for further development and utilization and quality evaluation of saffron resources.
