香蕉枯萎病菌‘热带4号’小种翻转酶家族的生物信息学分析

Bioinformatics Analysis of Flippase Family in Fusarium oxysporum f. sp. cubense ’Tropical Race 4’

摘要 由尖孢镰刀菌古巴专化型(Fusarium oxysporum f. sp. cubense, Foc)引起的香蕉枯萎病威胁着中国的香蕉产业,其中‘热带4号’生理小种(Foc tropical race 4, FocTR4)危害最大。胞外水解酶与效应蛋白是Foc重要的致病因子,它的分泌与膜运输系统密切相关,而翻转酶通过调控膜上的磷脂平衡来参与膜运输系统的调控。本研究以酿酒酵母(Saccharomyces cerevisiae)翻转酶家族氨基酸序列为材料,比对鉴定到FocTR4的翻转酶家族,对其进行生物信息学分析。结果表明:(1)通过蛋白进化树分析发现FocTR4具有5个翻转酶成员,分别位于4个亚组内,其中C亚组具有2个同源翻转酶。(2) FocTR4翻转酶家族C端磷脂转移酶功能域均为疏水性的,且相对保守。(3) FocTR4翻转酶家族除质膜定位以外,FocTR4DnfA与FocTR4DnfB还会定位于内质网,FocTR4DnfD还会定位于高尔基体。(4)在FocTR4和Foc1 (Foc race 1)翻转酶家族中,2个C亚组成员功能域区域发生了变异。本研究为进一步研究FocTR4翻转酶的生物学功能提供了科学依据,也为其它真菌翻转酶的研究提供了参考。 Banana fusarium wilt disease, caused by Fusarium oxysporum f. sp. cubense(Foc), especially the’Tropical Race 4’ of Foc(FocTR4), is threatening the banana industry in China. Foc can produce extracellular hydrolases and effector proteins as the important virulence-related agents, of which are associated with membrane trafficing system, and flippase can regulate membrane traffic by transporting phospholipids from the exoplasmic to the cytoplasmic leaflet to maintain the balance of phospholipid in membrane. We identified FocTR4 flippase family based on sequences of their homologous in Saccharomyces cerevisiae and analyzed their bioinformatics informations. Our results show that:(1) Flippase family of FocTR4 have 5 numbers, which are divided into 4 subgroups(A, B, C and D) and C-subgroup has two flippase members.(2) The PhoLip_ATPase_C domain of all FocTR4 flippases are hydrophobic and conservative.(3) Except plasma membrane localization of flippase family, FocTR4 DnfA and FocTR4 DnfB also localized to endoplasmic reticulum, and FocTR4 DnfD localized to Golgi.(4)Animo acid mutations exist in C-subgroup flipppases between FocTR4 and Foc1(Foc race 1). These results can provide scientific basis in further biological function researchs of flippase family in FocTR4, and as a reference to other fungal flippases’ study.
