摘要
【目的】研究小黄椰果实发育过程中椰子胚乳的抗氧化能力,为鲜食或加工用椰子果实的筛选提供理论参考。【方法】在小黄椰果实发育过程中取不同发育期椰子胚乳,分别测定椰子胚乳中的超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、谷胱甘肽还原酶(GR)活性及过氧化氢(H2O2)和可溶性蛋白含量,分析各指标的变化规律,评价不同成熟度椰子胚乳的抗氧化活性。【结果】液体胚乳的CAT、GR活性和H2O2含量高于固体胚乳,而SOD活性及可溶性蛋白含量低于固体胚乳。果实发育前期,固体胚乳的POD活性高于液体胚乳,而果实发育后期液体胚乳的POD活性高于固体胚乳。不同发育期中11月龄固体胚乳的SOD活性最强,为47.75 U/mgFW;而10月龄液体胚乳的POD活性最强,为8.41 U/mg;椰子胚乳中GR活力较低,最高仅为4.64 U/g,远低于的SOD、POD和CAT活性;6月龄液体胚乳的CAT活性最高,为8.75 U/mg。各抗氧化酶活性的相关性分析结果表明,椰子胚乳CAT活性与H2O2含量呈极显著正相关(P<0.01,下同),GR活性与可溶性蛋白含量呈显著负相关(P<0.05,下同);其中,液体胚乳的CAT活性与可溶性蛋白含量呈显著负相关,固体胚乳的CAT活性与GR活性呈显著正相关。【结论】椰子果实发育前期液体胚乳中的抗氧化酶起关键作用,在果实发育后期则固体胚乳的抗氧化酶逐渐起主导作用。因此,鲜食椰子可选择9月龄前的果实较好,而加工用椰子宜选择9月龄后的椰子果实。
【Objective】To study the antioxidant capacity of coconut endosperm during the development of small yellow coconuts,and to provide a theoretical reference for the screening of fresh or processed coconut fruits.【Method】During the development of small yellow coconut fruit,coconut endosperm at different developmental stages were taken,and the superoxide dismutase(SOD)activity and peroxidase(POD)activity,catalase(CAT)activity,glutathione reductase(GR)activity,hydrogen peroxide(H2 O2)content and soluble protein content in coconut endosperm were measured respectively,analysis of the changes in each index and evaluation of the coconut endosperm antioxidant activity at different maturity levels were conducted.【Result】The activities of CAT activity,GR activity and H2 O2 in liquid endosperm were higher than those in solid endosperm,while the SOD activity and soluble protein content were lower than those in solid endosperm.In the early stage of fruit development,the POD activity of solid endosperm was higher than that of liquid endosperm,while in the later stage of fruit development,the POD activity of liquid endosperm was higher than that of solid endosperm.The SOD activity of 11-month-old solid endosperm was the strongest at different developmental stages,which was 47.75 U/mgFW.The 10-month-old liquid endosperm had the strongest POD activity,at 8.41 U/mg;the GR activity in coconut endosperm was low,the highest one was only 4.64 U/g,which was far lower than the SOD,POD and CAT activities.The 6-month-old liquid endosperm had the highest CAT activity at 8.75 U/mg.The correlation analysis among various antioxidant enzymes activities showed that,there was extremely significant positive correlation between CAT activity and H2 O2 content of coconut endosperm(P<0.01,the same below),and there was significantly negative correlation between GR activity and soluble protein content(P<0.05,the same below).In liquid endosperm,the CAT activity had significant negative correlation with soluble protein content,and in solid endosperm,CAT activity had significant positive correlation with GR activity.【Conclusion】The antioxidant enzymes in the liquid endosperm play a key role in the early stage of coconut fruit development,and the antioxidant enzymes in the solid endosperm gradually play a leading role in the later stage of fruit development.Therefore,it is better to choose the fruit before 9 months of age for fresh coconut,and the coconut fruit after 9 months of age for processing coconut.
