摘要
目的:本研究旨在观察珍珠舌草胶囊全方挥发油对AGS细胞增殖和凋亡的作用,并对其机制进行研究。方法:依据2010版中国药典附录VD挥发油测定法甲法提取;给予不同浓度的药物干预AGS细胞的生长,采用MTT法测定细胞活力;应用流式细胞仪检测药物对胃癌AGS细胞周期和凋亡的影响;比色法测定药物对肿瘤细胞Caspase3活性的影响;药物干预期间用显微镜观察细胞数量及形态学变化。结果:珍珠舌草胶囊全方挥发油能呈浓度和时间依赖性地抑制胃癌细胞的活性,阻滞细胞周期于G 2/M期,促进胃癌细胞的凋亡,增强Caspase3的活性,药物干预期间细胞数量明显减少,部分细胞脱落、变圆,细胞壁边缘毛糙。结论:珍珠舌草胶囊全方挥发油具有抑制AGS细胞活性的作用,通过抑制细胞增殖和诱导细胞凋亡实现,促凋亡作用可能通过增强Caspase3的活性途径实现。
Objective:To study the effects and mechanism of the volatile oil of Zhenzhutongcao Capsule on the proliferation and apoptosis of gastric cancer cells.Methods:According to the appendix VD of Chinese Pharmacopoeia 2010,the volatile oil was extracted by method A.Different concentrations of drugs interfered with the growth of AGS cells.Cell viability was measured by MTT analysis.Cell cycle distribution was detected by flow cytometry(FCM);Caspase 3 activities were measured by colorimetric method;The changes of cell morphologic appearance after treatment were observed.Results:The volatile oil of zhenzhuzhicao capsule inhibited the activity of gastric cancer cells in a concentration and time-dependent manner,blocked the cell cycle in G 2/M phase,promoted the apoptosis of gastric cancer cells,and enhanced the activity of caspase 3.During the period of drug intervention,the number of cells decreased significantly.Some cells fell off and became round,and the cell wall edge was rough.Conclusions:The volatile oil of the whole prescription of zhenzhuzhicao capsule has the effect of inhibiting the activity of the AGS cells,which can be achieved by inhibiting cell proliferation and inducing apoptosis.The effect of promoting apoptosis may be achieved by enhancing the activity of caspase 3.
