摘要
为建立柞蚕微孢子虫体外培养增殖方法,找到适合微孢子虫增殖的昆虫细胞,将柞蚕微孢子虫接种High5、Sf21和柞蚕卵巢原代细胞后进行显微观察,分析不同昆虫细胞对柞蚕微孢子虫的敏感性。结果发现,柞蚕微孢子虫可以通过吞噬或胞饮作用进入High5和Sf21细胞中,但在这2种细胞的整个病变过程中,均未观察到孢子增殖阶段的其他形态;柞蚕微孢子虫接种柞蚕卵巢原代细胞后,在细胞病变过程中可以观察到裂殖体、产孢体、孢子母细胞等增殖阶段的孢子形态,后期扩繁的孢子大量形成并释放。结果表明,柞蚕微孢子虫在High5、Sf21这2种细胞中不能扩繁增殖,但可以侵染柞蚕卵巢原代细胞,并大量增殖扩繁。
In order to develop appropriate method for in vitro propagation of Nosema pernyi,insect cell lines High5 and Sf21,and primary ovarian cell from Antheraea pernyi were inoculated with N. pernyi spores to test their susceptibility by microscopy. Results showed that spores of N. pernyi could gain access to High5 and Sf21 cells by endocytosis and phagocytosis. But no other proliferative phases could be observed during the whole cytopathic process of High5 and Sf21 cells. When A. pernyi primary ovarian cells were incubated with N. pernyi,proliferative phases including schizont,sporont,sporoblast and final release of proliferated spores could be observed during the cytopathic process. In conclusion,N. pernyi could not propagate in heterologous cell lines High5 and Sf21,but could infect A. pernyi primary ovarian cell and numerously propagate.
