籽鹅羽束性状在肝脏中相关候选基因关联性分析

Association analysis of related candidate genes of liver for crest trait in zi-goose

摘要 本研究旨在分析籽鹅头顶部羽束性状(crest trait)相关调控基因,从而进一步揭示禽类表型性状与调控基因的作用机制,为后续分子遗传育种、禽类基因编辑等提供理论基础。本试验选取同家系同世代1日龄籽鹅作为试验样本,构建2个资源群体,每群体60只雏鹅,饲养周期为21 d。有羽束鹅群(crest population,C):该群体均为有羽束公雏;无羽束鹅群(non-crest,NC):该群体均为无羽束公雏,2个群体同舍分栏饲养。随后,为测定肝脏组织中eomes、hoxc8、sox5、mnr2、wnt5a、bmp2、gh候选基因相对表达量,在1,7,14,21日龄进行采样,应用qRT-PCR技术进行检测。结合生物信息学分析方法,分析调控羽束性状的关联基因。在qRT-PCR分析结果中,sox5基因在1日龄雏鹅肝脏组织中相对表达量,C组高于NC组,并且差异极显著(P<0.01)。其他时期虽无显著差异,但羽束鹅群仍高于无羽束鹅群相对表达量高。wnt5a基因在1日龄雏鹅肝脏中相对表达量,C组高于NC组,差异极显著(P<0.01),在其他时期无显著差异。通过生物信息学分析,结合Multiexperiment Viewer软件对候选基因进行聚类分析,sox5、wnt5a、bmp2基因可能均在同一调控通路中。结果提示,sox5基因可能为羽束性状调控基因,与wnt5a基因在同一信号通路中,并对羽束性状发育起到调控作用,sox5、bmp2基因可能在肝脏发育、代谢过程中起到调控作用。 This experiment was conducted to study the related regulation genes of the crest trait on the top of the head of the goose,and so as to further reveal the mechanism of regulation of gene and phenotypic traits of poultry,and provide a theoretical basis and reference for molecular genetic breeding,poultry gene editing etc..In this study,1 day old geese were selected as experimental samples,and two resource populations were established,60 geese per group,and the feeding period was 21 days,including groupⅠ(crest population,C):the male geese with crest trait,and groupⅡ(non-crest,NC):the male geese with non-crest.Two experimental group was reared in the same birdhouse.Subsequently,the relative expression level of the candidate genes of eomes,hoxc8,sox5,mnr2,wnt5 a,bmp2,gh in liver was detected at 1,7,14,21 days old after hatching by qRT-PCR,screened candidate genes for crest trait and analyzed the association by bioinformatics.In the result of qRT-PCR analysis,the relative expression of sox5 in the liver tissue of crest trait male at 1 day old was significant higher than that of non-crest(P< 0.01).Although the crest traits male still had higher gene expression than the non-crest traits,the differences were not significant at other age days.The relative expression of wnt5 a in the liver tissue of crest trait male at 1 day old was significant higher than that of non-crest(P<0.01),and there was no significant at other days.The candidate genes were clustered by Multiexperiment Viewer tool.Candidate genes of sox5,wnt5 a,bmp2 may be in the same regulatory pathway.These results indicated that sox5 gene may be a gene of regulating crest traits.It and wnt5 agene are in the same signal pathway in regulation of the development of crest traits.ox5 and bmp2 genes may play a regulatory role in the development and metabolism of the liver.
