阿替普酶对脑梗死大鼠的疗效观察及对血清中GAP-43、NGF、MDA水平的影响 被引量:4

摘要 目的研究阿替普酶对脑梗死大鼠的疗效及对血清中生长相关蛋白-43(GAP-43)、神经生长因子(NGF)、丙二醛(MDA)水平的影响。方法选择SD健康雌性大鼠30只,20只大鼠进行脑梗死模型制备,建模成功共16只,将建模成功的大鼠随机分为模型组和阿替普酶组,每组8只,正常组大鼠10只。阿替普酶组大鼠静脉注射阿替普酶,5 mg/kg;正常组和模型组大鼠注射等体积的无菌0.9%氯化钠溶液。脑梗死体积TTC染色,脑组织含水量测定,大鼠ZeaLonga评分,在显微镜下观察大鼠脑组织病理学特征。酶联免疫吸附法(ELISA)检测GAP-43、NGF、MDA水平,Western blot检测PTEN、mTOR、PKA蛋白表达。结果模型组大鼠脑梗死体积、脑组织含水量、ZeaLonga评分显著高于正常组和阿替普酶组,差异有统计学意义(P<0.05);阿替普酶组大鼠脑梗死体积、脑组织含水量、ZeaLonga评分低于模型组,差异有统计学意义(P<0.05)。模型组GAP-43、NGF、MDA水平显著高于正常组和阿替普酶组,差异有统计学意义(P<0.05);阿替普酶组GAP-43、NGF、MDA水平低于模型组,差异有统计学意义(P<0.05)。模型组PTEN、mTOR、PKA表达水平高于正常组和阿替普酶组,差异有统计学意义(P<0.05);阿替普酶组PTEN、mTOR、PKA表达水平低于模型组,差异有统计学意义(P<0.05)。结论阿替普酶对脑梗死大鼠的治疗效果显著,促进大鼠神经功能恢复,对GAP-43、NGF、MDA水平有抑制作用。 Objective To investigate the effects of alteplase on the serum levels of growth associated protein-43(GAP-43),nerve growth factor(NGF)and malondialdehyde(MDA)in rats with cerebral infarction.Methods Thirty SD healthy female rats were enrolled in the study,of whom,20 SD healthy female rats were selected to establish cerebral infarction models,and 16 rat models with cerebral infarction were successfully established,then the rats were randomly divided into model group and alteplase group,with 8 rats in each group,moreover the other 10 rats were served as control group.The rats in alteplase group were intravenously injected with alteplase 5mg/kg,and the rats in control group injected with the same volume of sterile saline.The volume of cerebral infarction was stained by TTC,the water content of brain tissue was measured,and the Zea Longa scores were observed.ELISA method was used to detect the levels of GAP-43,NGF and MDA.Western blot was used to detect the expression levels of PTEN,mTOR and PKA protein.Results The volume of cerebral infarction,water content of brain tissue and Zea Longa scores in model group were significantly higher than those in control group and alteplase group(P<0.05),and the volume of cerebral infarction,water content of brain tissue and Zea Longa scores in alteplase group were significantly lower than those in control group(P<0.05).The levels of GAP-43,NGF and MDA in model group were significantly higher than those in control group and in alteplase group(P<0.05),however,the levels of GAP-43,NGF and MDA in alteplase group were significantly lower than those in model group(P<0.05).The expression levels of PTEN,mTOR and PKA in model group were significantly higher than those in control group and alteplase group(P<0.05),and the expression levels of PTEN,mTOR and PKA in alteplase group were significantly lower than those in model group(P<0.05).Conclusion The alteplase has significant therapeutic effects on cerebral infarction in rat models,which can promote the recovery of neural function,and inhibits the expression levels of GAP-43,NGF and MDA in rats with cerebral infarction.
