摘要
目的探讨顺铂(DDP)耐药宫颈癌细胞中沉默交配型信息调节因子2同源蛋白1(SIRT1)的表达及其对细胞增殖的影响。方法将宫颈癌HeLa和SiHa细胞分别分为耐药组和敏感组。敏感组细胞不做任何处理;耐药组细胞在培养过程中逐步增加培养液中顺铂的浓度,建立对对顺铂耐药的HeLa/DDP和SiHa/DDP细胞系。再将HeLa/DDP和SiHa/DDP细胞系分为HeLa/DDP-NC组、HeLa/DDP-siRNA组以及SiHa/DDP-NC组、SiHa/DDP-siRNA组,并分别转染相应的siRNA。检测耐药组和敏感组细胞SIRT1蛋白及mRNA表达水平;检测DDP-NC组与DDP-siRNA组细胞SIRT1蛋白及mRNA表达水平及增殖情况。结果耐药组HeLa及SiHa细胞SIRT1蛋白及mRNA表达水平均高于敏感组(均P<0.05)。DDP-NC组HeLa及SiHa细胞增殖水平高于DDP-siRNA组(均P<0.05)。结论顺铂耐药宫颈癌细胞SIRT1表达水平升高,抑制SIRT1表达可以抑制耐药细胞的增殖。
Objective To explore the expression of silent mating-type information regulator 2 homolog 1(SIRT1)and its effect on cell proliferation in cisplatin(DDP)-resistant cervical cancer cells.Methods Cervical cancer HeLa/SiHa cells were divided into resistance group and sensitive group.The cells in the sensitive group did not receive any treatment,whereas the cells in the resistance group received cisplatin at a gradually increased concentration in the culture medium during culture to establish cisplatin-resistant HeLa/DDP,SiHa/DDP cell lines.Furthermore,HeLa/DDP and SiHa/DDP cell lines were divided into HeLa/DDP-NC group,HeLa/DDP-siRNA group,SiHa/DDP-NC group and SiHa/DDP-siRNA group,and separately received transfection with corresponding siRNAs.The expression levels of SIRT1 protein and mRNA were detected in cells of the resistance and sensitive groups,the expression levels of SIRT1 protein and mRNA and proliferation were detected in the cells of the DDP-NC and DDP-siRNA groups.Results The expression levels of SIRT1 protein and mRNA in HeLa/SiHa cells of the resistance group were higher than those of the sensitive group(all P<0.05).The proliferation level of HeLa/SiHa cells in the DDP-NC group was higher than that in the DDP-siRNA group(all P<0.05).Conclusion Cisplatin-resistant cervical cancer cells exhibit elevated SIRT1 expression level,and inhibiting SIRT1 expression can suppress the proliferation of resistant cells.
