摘要
目的探讨儿茶素提取物表没食子儿茶素没食子酸酯(EGCG)对牙龈卟啉单胞菌的体外抑菌活性。方法检测EGCG对牙龈卟啉单胞菌的最小抑菌浓度值(MIC)及最小杀菌浓度(MBC)。配制浓度分别为1/4 MIC值、1/2 MIC值、MIC值的EGCG溶液,检测其对牙龈卟啉单胞菌生长的影响,并绘制生长曲线。配制浓度分别为1/2 MIC、MIC、2 MIC、4 MIC、8 MIC、16 MIC的EGCG,检测不同浓度EGCG对牙龈卟啉单胞菌生物膜的抑制率,并计算最低清除浓度(MBRC) 50。结果 EGCG对牙龈卟啉单胞菌的MIC值为0. 250 0 mol/L,MBC值为1. 000 0 mol/L。各实验组在0~12 h为生长迟缓期,12 h后进入对数生长期,0. 250 0 mol/L EGCG组在24 h后进入平台期,而0. 125 0 mol/L EGCG组、0. 062 5 mol/L EGCG组则处于缓慢增长状态。MBRC50为4. 0 mol/L。EGCG浓度分别为0. 5、1. 0、2. 0、4. 0 mol/L时对P. gingivalis生物膜的抑制率组间两两比较,差异均有统计学意义(P <0. 05)结论儿茶素提取物EGCG是一种安全、无耐药性的天然药物,体外研究证实其对牙龈卟啉单胞菌有明显的抑制作用。
Objective To investigate the antibacterial activity of epigallocatechingallate( EGCG) extract against Porphyromonasgingivalis( P. gingivalis) in vitro. Methods The minimum inhibitory concentration( MIC) and minimum bactericidal concentration( MBC) of EGCG on P. gingivalis were determined. EGCG solutions with concentrations of 1/4 MIC,1/2 MIC and MIC were prepared to detect the effects on the growth of P. gingivalis,and the growth curve was drawn. EGCG with concentrations of 1/2 MIC MIC,2 MIC,4 MIC,8 MIC and 16 MIC were prepared to detect the inhibition rate of different concentrations of EGCG on P. gingivalis biofilm,and minimum biofilm reduction concentration( MBRC) 50 was calculated. Results The MIC of EGCG to P. gingivalis was 0. 250 0 mol/L,the MBC of EGCG to P. gingivalis was 1. 000 0 mol/L. The lag phase was from 0 hour to 12 hours and the logarithmic growth phase was after 12 hours in each experimental group. The group of 0. 250 0 mol/L of EGCG entered plateau phase after 24 hours,but the group of 0. 125 0 mol/L and 0. 0625 mol/L of EGCG entered slow growth phase after 24 hours. MBRC50 was 4. 0 mol/L. There were statistically significant differences in the inhibitory rate in each experimental group( P < 0. 05). Conclusion EGCG as a safe,no drug resistance and natural medicine has proved the significant inhibition to Porphyromonasgingivalis in vitro.
