摘要
目的:优化超声波辅助提取7种地黄成方制剂中含量测定工艺。方法:以马钱苷和丹皮酚为考察指标,通过单因素试验考察超声波功率、超声波频率、提取容器、提取溶剂、提取时间、料液比、提取温度,采用L16(45)正交试验设计确定最佳提取工艺,对市场不同厂家成方制剂进行测定。结果:超声波功率在250~600 W,超声波频率在25~60 KHz提取效果比较,差异无统计学意义(P>0.05);采用玻璃容器提取,影响超声波辅助提取的主次因素为提取时间>料液比>提取溶剂>提取温度,最佳提取工艺提取时间为30 min、料液比为1∶100、提取溶剂为45%甲醇、提取温度为50℃。除六味地黄软胶囊外,其他6种超声波辅助提取均优于传统加热提取。结论:超声波辅助提取能显著缩短提取时间、简化实验操作,可用于马钱苷和丹皮酚的含量测定。
Objective:To optimize the ultrasonic-assisted extraction process for the determination of seven preparations of Radix Rehmanniae.Methods:Taking loganin and paeonol as indexes,the ultrasonic power,frequency,extraction container,extraction solvent,extraction time,Solid-liquid ratio and extraction temperature were investigated by single factor experiment.The optimum extraction process was determined by L16(45)orthogonal design.And the preparations from different manufacturers were determined.Results:There was no significant difference in the extraction effect(P>0.05)with the ultrasonic power of 250-600 W and the ultrasonic frequency of 25-60 KHz.Extraction by glass container,the effectiveness of factors on extraction was in this order:extraction time,Solid-liquid ratio,extraction solvent,extraction temperature.The optimized extraction conditions are as follows:extraction time with 30 min,Solid-liquid ratio 1∶100,45%methanol,extraction temperature with 50℃.Ultrasonic-assisted extraction were superior to traditional heating extraction except Liuwei Dihuang Soft Capsule(六味地黄软胶囊).Conclusion:Ultrasound-assisted extraction can significantly shorten the extraction time and simplify the experimental operation.It can be used for the determination of loganin and paeonol.
