RP-HPLC法测定解毒饮中有机酸类和黄酮类成分 被引量:2

摘要 目的:建立同时测定解毒饮中新绿原酸、隐绿原酸、绿原酸、咖啡酸、异绿原酸A、异绿原酸B、异绿原酸C、黄芩苷、汉黄芩苷9种有机酸类和黄酮类成分含量的方法。方法:采用RP-HPLC法,色谱柱:Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm);流动相:乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱:0~10 min,8 A%;10~25 min,8~15 A%;25~40 min,15~20 A%;40~50 min,20~30 A%;50~60 min,30~50 A%;流速:1 mL/min;柱温:40℃;检测波长:280/325 nm;进样量:10μL。结果:新绿原酸、绿原酸、隐绿原酸、咖啡酸、异绿原酸A、异绿原酸B、异绿原酸C、黄芩苷、汉黄芩苷9种有效成分别在1.62~16.18、8.69~86.86、2.19~21.88、0.21~2.12、1.83~18.26、1.14~11.42、1.87~18.68、14.45~144.52、3.49~34.86μg/mL范围内呈良好的线性关系(r≥0.999),平均回收率分别为98.36%、99.96%、99.48%、101.30%、101.42%、99.72%、100.61%、100.09%、99.87%,RSD均<2.00%。结论:所建HPLC分析方法简单、准确可靠、专属性强、重现性良好,可应用于解毒饮中有机酸及黄酮类成分的含量测定。 Objective:To establish a method for the determination of nine organic acids and flavonoids concerning neochlorogenic acid,cryptochlorogenic acid,chlorogenic acid,caffeic acid,isochlorogenic acid A,isochlorogenic acid B,isochlorogenic acid C,baicalin and wogonoside in Jiedu decotion.Methods:RP-HPLC was adopted in this study;Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm)was applied to separate the sample at the column temperature of 40℃.Acetonitrile(A)-0.1%H3PO4 aqueous solution(B)was used as the mobile phase.Gradient elution method:0-10 min,8 A%;10-25 min,8-15 A%;25-40 min,15-20 A%;40-50 min,20-30 A%;50-60 min,30-50 A%.The flow rate was 1 mL/min;the detection wavelength was 280 and 325 nm.The injection volume was 10μL.Results:The linear range of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,caffeic acid,isochlorogenic acid A,isochlorogenic acid B,isochlorogenic acid C,baicalin and wogonoside were respectively 1.62-16.18,8.69-86.86,2.19-21.88,0.21-2.12,1.83-18.26,1.14-11.42,1.87-18.68,14.45-144.52,3.49-34.86μg/mL(r≥0.999)which indicated the linearity of this method is satisfactory.The average recovery of the determined compounds were respectively 98.36%,99.96%,99.48%,101.30%,101.42%,99.72%,100.61%,100.09%,99.87%,and the RSD<2.00%.Conclusion:This method is simple,accurate,specific,reliable and has good reproducibility,which can be used for the determination of organic acids and flavonoids in Jiedu decotion.
