摘要
目的:探讨二甲双胍联合S-烯丙基巯基半胱氨酸(S-allylmercaptocysteine,SAMC)通过自噬改善糖尿病小鼠肝脏胰岛素抵抗的实验效果。方法:随机选取40只C57BL/6J小鼠,通过高饲喂养后分别体内注射小剂量链脲佐菌素(STZ),建立糖尿病小鼠胰岛素抵抗模型。而后随机分为四组,分别为对照组、二甲双胍组、SAMC组、二甲双胍联合SAMC组,通过四种不同的处理方式,4周后,分析四组小鼠胰岛素、C肽、血糖、胰高血糖素、瘦素以及肿瘤坏死因子等指标的变化情况。结果:二甲双胍联合SAMC组小鼠的胰岛素、C肽、血糖、胰高血糖素、瘦素以及肿瘤坏死因子等多种生化指标水平明显低于其他三组,差异有统计学意义(P<0.05)。而对照组、二甲双胍组以及SAMC组三组C57BL/6J糖尿病小鼠的生化指标差异不显著(P>0.05)。结论:二甲双胍联合SAMC通过自噬改善糖尿病小鼠胰岛素抵抗的状态,对降低糖尿病小鼠的血糖具有显著疗效。而单纯使用二甲双胍或者单纯使用SAMC对降低糖尿病小鼠的血糖以及改善胰岛素抵抗状态作用不明显。
Objective:To investigate the effect of metformin combined with S-allylmercaptocysteine(SAMC)on improving liver insulin resistance in diabetic mice by autophagy.Methods:40 C57BL/6J mice were randomly selected and injected with small dose of streptozotocin(STZ)in vivo after high feeding to establish insulin resistance model of diabetic mice.Then they were randomly divided into four groups:control group,metformin group,SAMC group,metformin combined with SAMC group.After 4 weeks,the changes of insulin,C-peptide,blood sugar,glucagon,leptin and tumor necrosis factor were analyzed.Results:The levels of insulin,C-peptide,blood glucose,glucagon,leptin and TNF in metformin combined with SAMC group were significantly lower than those in other three groups(P<0.05).The biochemical indexes of C57BL/6J diabetic mice in control group,metformin group and SAMC group were not significantly different(P>0.05).Conclusion:Metformin combined with SAMC can improve insulin resistance in diabetic mice by autophagy,and has significant effect on reducing blood sugar in diabetic mice.Metformin alone or SAMC alone had no significant effect on reducing blood sugar and improving insulin resistance in diabetic mice.
