摘要
为利用响应面法优化金银花叶总黄酮有效部位的制备工艺,并考察其体外抗氧化能力,试验对4种型号大孔树脂的吸附-解吸能力进行比较,并筛选最优的大孔树脂型号;采用单因素试验和Box-Behnken响应面法优化大孔树脂纯化金银花叶总黄酮有效部位的制备工艺,分别考察洗脱溶剂浓度、洗脱溶剂体积、上样流速、洗脱流速等因素水平,并进行三次验证试验;以抗坏血酸为阳性对照,测定金银花叶黄酮有效部位的DPPH自由基清除能力和ABTS+自由基的清除能力。结果表明:优选D101大孔树脂最佳纯化工艺为:上样流速2.5 mL/min,洗脱溶剂为60%乙醇,洗脱溶剂体积为4倍柱体积,洗脱流速2 mL/min;制备的有效部位提取物中总黄酮的含量超过50%;60%乙醇部位体外抗氧化能力试验证明金银花叶总黄酮有效部位具有良好的体外抗氧化能力。采用Box-Behnken响应面法优化的大孔树脂纯化金银花叶总黄酮有效部位的工艺可行,且稳定性好;金银花叶总黄酮有效部位的体外抗氧化效果显著,值得进一步研究开发。
To optimize the preparation process of the effective fraction of total flavonoids from honeysuckle leaves by response surface methodology,and to investigate its antioxidant capacity in vitro.The adsorption-desorption capacity of four types of macroporous resins was compared and the optimal macroporous resin model was screened.The single-factor test and Box-Behnken response surface method were used to optimize the effective fraction of total flavonoids in honeysuckle leaves.The preparation process was carried out to investigate the factors such as elution solvent concentration,elution solvent volume,loading flow rate and elution flow rate,and three verification tests were carried out.Ascorbic acid was used as positive control to determine DPPH free radical scavenging ability of the effective fraction of flavonoids in honeysuckle leaves.And ABTS+free radical scavenging ability.Results showed that:The D101 macroporous resin was optimized for purification.The optimal purification process was as follows:the elution solvent was 60%ethanol,the elution solvent volume was 4 column volumes,the loading flow rate was 2.5 mL/min,and the elution flow rate was 2 mL/min.The content of total flavonoids in the prepared extracts of the extracts exceeded 50%;the antioxidant activity in vitro showed that the effective fraction of total flavonoids of honeysuckle leaves had good antioxidant capacity in vitro.The macroporous resin optimized by Box-Behnken response surface method was effective in purifying the effective fraction of total flavonoids from honeysuckle leaves,and the stability was good.The anti-oxidation effect of the effective fraction of total flavonoids in honeysuckle leaves was significant,which was worthy of further research and development.
