摘要
目的探讨当归补血汤干预肺纤维化大鼠气虚血瘀状态的机制。方法选取90只雄性SD大鼠随机分为假手术组、模型组和当归补血汤组,每组30只。博莱霉素气管内滴注法诱导肺纤维化大鼠模型。手术后第2天,假手术组和模型组灌胃生理盐水10 ml/(kg·d),当归补血汤组灌胃中药煎液3.75 g/(kg·d)。分别于实验第7、14和28天每组随机抽取10只大鼠,观察体质量变化,并评估气虚血瘀证候量化评分,行负重游泳实验后断头取血、取肺,血标本经抗凝后检测血液流变学指标,肺组织行HE和Masson染色观察。结果与假手术组比较,模型组各时间节点体质量和负重游泳时间明显下降,气虚血瘀证侯量化评分和全血黏度明显升高(P<0.05或P<0.01)。当归补血汤可显著改善大鼠的体质量,提高负重游泳时间,下调气虚血瘀证候量化评分和全血黏度(P<0.05或P<0.01)。结论当归补血汤可通过调节血液流变学指标,改善肺纤维化大鼠气虚血瘀状态。
Objective To investigate the mechanism of Danggui Buxue Decoction for intervention of Qi deficiency and blood stasis in rats with pulmonary fibrosis.Methods Ninety male male SD rats were randomly divided into sham operation group,model group and Danggui Buxue decoction group,with 30 rats in each group.The rat model of pulmonary fibrosis was induced by intratracheal instillation of bleomycin.On the second day after operation,the sham operation group and the model group were perfused with saline 10 ml/(kg·d),and the Danggui Buxue Decoction group was perfused with 3.75 g/(kg·d)of Chinese medicine decoction.Ten rats were randomly selected from each group at 7,14 and 28 d after the experiment to observe the changes of body weight and evaluate the quantified scores of Qi deficiency and blood stasis syndrome.After the weight-bearing swimming test,the blood was taken after decapitation and the lungs were collected.The blood samples were tested for hemorheological changes after anticoagulation,and the lung tissues were observed by HE and Masson staining.Results Compared with the sham operation group,the body weight and weight-bearing swimming time at each time point in the model group were significantly reduced,whereas the scores of Qi deficiency and blood stasis syndrome and whole blood viscosity were significantly increased(P<0.05 or P<0.01).Danggui Buxue decoction significantly improved the body weight change,increased the weight-bearing swimming time,down-regulated the scores of Qi deficiency and blood stasis syndrome and the whole blood viscosity(P<0.05 or P<0.01).Conclusion Danggui Buxue decoction can improve the state of Qi deficiency and blood stasis in rats with pulmonary fibrosis by regulating hemorheology indexes.
