摘要
目的探讨长链非编码肺腺癌转移相关转录子1(long non-coding metastasis-associated lung adenocarcinoma transcript 1,MALAT1)在肝癌中的表达情况及其对化疗药物敏感性的影响。方法采用实时荧光定量PCR检测92例肝癌组织及其对应癌旁正常组织中lnc-MALAT1的表达量;分析lnc-MALAT1表达量与肝癌患者临床特征和总生存期的关系。使用HpG2细胞转染MALAT1过表达质粒,转染空载体作为对照组。通过CCK-8实验测定过表达lnc-MALAT1后对细胞增殖的影响;通过化疗药物敏感性实验测定过表达lnc-MALAT1后细胞对化疗药物的敏感性影响。结果lnc-MALAT1在肝癌组织中高表达(P<0.01);其高表达在肝癌患者临床分期(P=0.002)、原发病灶(P=0.027)、淋巴结转移(P=0.026)、远端转移(P=0.004)、化疗(P=0.012)及乙肝表面抗原(hepatitis B surface antigen,HBsAg;P=0.039)方面比较,差异均具有统计学意义。高表达患者总生存时间短于低表达患者[(26.69±16.02)个月vs(37.14±15.77)个月,P=0.027]。转染48 h后,过表达lnc-MALAT1能够促进HpG2细胞增殖(P<0.05)。使用≥0.01 ng/mL顺铂和≥0.1 ng/mL氟尿嘧啶均发现过表达MALAT1能够降低HpG2细胞对以上2种药物的敏感性(均P<0.01)。结论lnc-MALAT1在肝癌中高表达,能够缩短患者生存期,并降低肝癌细胞对化疗药物的敏感性。
Objective To detect the expression pattern of long non-coding metastasis-associated lung adenocarcinoma transcript 1(lnc-MALAT1)in liver carcinoma and its effect on the sensitivity of chemotherapy.Methods Real-time quantitative PCR was used to detect the expression of lnc-MALAT1 in liver cancer tissues and normal tissues adjacent to the cancer.The relationship between the expression of lnc-MALAT1 and the clinical characteristics and overall survival of liver cancer patients were analyzed.HpG2 cells were used to transfect the MALAT1 overexpression plasmid,and the empty vector was used as a control group.The effect of overexpression of lnc-MALAT1 on cell proliferation was measured by CCK-8 experiment,and the sensitivity of chemotherapy after overexpression of lnc-MALAT1 on HpG2 cells was determined by chemotherapeutic drug sensitivity experiment.Results lnc-MALAT1 was highly expressed in liver cancer tissues(P<0.01);liver cancer patients with an overexpression of lnc-MALAT1 had a shorter overall survival[(26.69±16.02)months vs(37.14±15.77)months,P=0.027].The highly expressed lnc-MALAT1 was correlated with primary lesions(P=0.027),lymph node metastasis(P=0.002),distant metastasis(P=0.004),chemotherapy(P=0.012),and hepatitis B surface antigen(HBsAg,P=0.039)of liver cancer patients.48 h after transfection of HpG2 cells,the overexpression of lnc-MALAT1 promoted the proliferation(P<0.05)and inhibited the sensitivity of chemotherapy(≥0.01 ng/mL cisplatin or≥0.1 ng/mL 5-FU,all P<0.01).Conclusions lnc-MALAT1 is highly expressed in liver cancer tissues,which can shorten the overall survival of liver cancer patients,and reduce the sensitivity of liver cancer cells to chemotherapy drugs.
