摘要
目的建立紫外分光光度法测定莲子心中总黄酮的含量。方法采用超声波辅助乙醇提取莲子心,经亚硝酸钠-硝酸铝-氢氧化钠显色,以芦丁为对照品,利用采用紫外分光光度计在500nm波长处测定。结果莲子心中总黄酮平均含量为1.13%,加标回收率在96.65%~98.48%,RSD在0.65%~1.7%。结论该法操作简单、准确、重现性好,可为莲子心药材质量控制提供方法。
OBJECTIVE To established a method of determination of total flavonoids in lotus seeds by ultraviolet spectrophotometry.METHODS The sample was extracted by ultrasound-assisted ethanol,dyed by sodium nitrite-aluminum nitrate-sodium hydroxide,used rutin as reference substance,determined by ultraviolet spectrophotometer at 500 nm.RESULTS The total flavonoid content in lotus seeds was 1.13%,the recovery rate was in the range of 96.65%-98.48%,and the RSD was in the range of 0.65%-1.7%.CONCLUSION The UPLC-MS/MS method has high accuracy,high sensitivity and good reproducibility.
