肝豆灵对高铜负荷大鼠空间学习记忆能力及神经干细胞ROS水平的影响 被引量:3

摘要 目的:观察肝豆灵对高铜负荷大鼠空间学习记忆能力及胎鼠神经干细胞(neural stem cell,NSC)活性氧(reactive oxygen species,ROS)水平的影响。方法:75只SD大鼠适应性喂养7d,随机分为对照组,模型组,肝豆灵处理高、中、低剂量组,肝豆灵处理组分别给予1.92 g/kg/d、0.96 g/kg/d、0.48 g/kg/d药物血清灌胃,对照组以等体积纯净水灌胃,7d后观察SD大鼠在T臂迷宫试验中空间学习记忆能力的变化。于无菌条件下取孕14d ICR小鼠胎鼠海马组织,分为对照组、高铜模型组和高、中、低剂量肝豆灵片处理组,对照组在液体培养基中加入10%的SD大鼠空白血清,模型组用含10%的SD大鼠空白血清铜负荷培养基培养,肝豆灵低、中、高剂量组予以3种不同剂量配制的10%的肝豆灵药物血清铜负荷培养基培养,以流氏细胞仪检测神经干细胞ROS水平。结果:行为学试验,模型组和小剂量治疗组的进入电刺激区的时间明显降低,经过药物治疗后,进入电刺激区的时间(S)明显增加。ROS水平检测,模型组高于对照组(P<0.01),肝豆灵组低于模型组(P<0.05),高于正常组(P<0.05)。结论:化痰祛瘀方肝豆灵可降低神经干细胞中ROS的水平并改善高铜负荷大鼠空间学习记忆能力。 Objective To observe the effects of Gandouling on spatial learning and memory in rats with high copper load and the effects of Gandouling on ROS levels in fetal neural stem cells.Method Adaptability of 75 SD rats fed,after 7 days,were randomly divided into control group,model group,hepatic bean spirit high,medium and low dose group,treatment group were given liver beans spirit of 1.92 g/kg/d,0.96 g/kg/d,0.48 g/kg/d drug serum to fill the stomach,the control group with pure water to fill the stomach,such as volume seven days to observe SD rats in T arm maze test the change of spatial learning and memory ability.In a sterile conditions get pregnant 14 d fetal rat hippocampus,ICR mice were divided into control group,high copper model group and high,medium and low dose hepatic bean spirit treatment group,control group to join in the liquid medium blank serum,10%of the SD rats model group with blank SD rats serum copper containing 10%load medium,hepatic bean spirit high,medium and low dose group to be three different dose makes up 10%of the cultured liver beans spirit drug serum copper load medium,flow meter’s cells in detecting neural stem cells ROS levels.Result In the behavioral test,the time to enter the electrical stimulation area in the model group and the low-dose treatment group decreased significantly,and the time to enter the electrical stimulation area(S)increased significantly after drug treatment.ROS level in the model group was higher than that in the control group(P<0.01),lower than that in the model group(P<0.05),and higher than that in the normal group(P<0.05).Conclusion Gandouling Decoction for Resolving Phlegm and Removing Stasis can reduce ROS level in neural stem cells and improve spatial learning and memory ability of rats with high copper load.
