摘要
以凤梨叶片为筛选材料,通过平板对峙培养法筛选出3株对蔬菜软腐病菌菊迪基氏菌有拮抗效果的生防细菌,结合生理生化及分子生物学鉴定,以及拮抗效果评价试验,结果表明,这3株内生菌分别为:Bacillus amyloliquefaciens F-2,Bacillus megaterium F-3-1,Bacillus subtilis F-3-2,皆为革兰氏阳性菌.其中F-2对菊迪基氏菌和丁香假单胞菌的抑菌效果最好,抑菌圈分别为24.67 mm和28.83 mm;F-3-1对病原真菌燕麦镰孢菌、胶孢炭疽菌、互隔交链孢霉,抑菌率分别为70.83%、91.67%、65.56%.F-3-2对丁香假单胞菌和黄单胞菌属的抑菌效果最好,抑菌圈分别为20.75 mm和20.00 mm.另外,通过生化鉴定研究发现,这3株优良内生菌还具有很好的解磷固氮和产纤维素酶能力,这对内生菌的筛选及内生菌对植物抗病促生作用的研究提供了一定的参考价值.
Using pineapple leaves as screening material,three strains of biocontrol bacteria with antagonistic effect on Dickeya chrysanthemi were screened by plate confrontation method.Combined with the physiological and biochemical and molecular biology identification,the results showed that the three strains endophytes were:Bacillus amyloliquefaciens F-2,Bacillus megaterium F-3-1 and Bacillus subtilis F-3-2,which were all gram positive bacteria.Evaluation test of antagonistic effect showed that F-2 had the best bacteriostatic effect on D.chrysanthemi and Pseudomonas syringae,with the bacteriostatic zone of 24.67 mm and 28.83 mm.The inhibit rates of F-3-1 against Fusarium avenaceum,Colletotrichum gloeosporioides and Alternaria alternate zzha were 70.83%,91.67%,and 65.56%,respectively.F-3-2 had the best bacteriostatic effect on P.syringae and Xanthomonas arboricola,with a bacteriostatic zone of 20.75 mm and 20.00 mm.In addition,through biochemical identification,it was found that these three endophytic bacteria also had good ability of dissolved phosphorus,nitrogen fixation and cellulase production.This work provided a reference for the screening of endophytic bacteria and the study on the role of endophytic bacteria in plant disease resistance and growth promotion.
