摘要
目的:探究羟基喜树碱(HCTP)联合人类细胞因子诱导的杀伤细胞(CIK)对肝癌细胞增殖的抑制作用。方法:以体外培养的肝癌BEL-7402细胞株为实验研究对象,将HCPT设9个浓度组,CIK细胞设5个效靶比组,分别干预肝癌BEL-7402细胞24 h、48 h及72 h后,MTT法检测HCPT和CIK细胞对肝癌细胞的抑制作用。筛选HCPT的最佳浓度和CIK细胞的最佳效靶比,联合干预肝癌BEL-7402细胞24 h、48 h和72 h后,Annexin V/PI双染色法检测细胞凋亡。结果:HCPT(最佳浓度0.5 mg/L)和CIK细胞(最佳效靶比20∶1)在不同时间对肝癌细胞BEL-7402均有生长抑制作用,而联合应用具有协同作用,可更好发挥抗癌作用,比较差异有统计学意义(P<0.05)。结论:HCPT和CIK细胞治疗对肝癌BEL-7402细胞具有较明显的抑制作用,两者联合作用有协同作用,并且能够诱导细胞凋亡。
Objective To investigate the inhibitory effect of hydroxycamptothecin(HCTP)combined with human cytokine-induced killer cells(CIK)on the proliferation of hepatoma cells.Method The human hepatoma cell line BEL-7402 was cultured in vitro.The HCPT was set in 9 concentration groups and the CIK cells were set in 5 effective target ratio groups.The cells were treated with BEL-7402 cells for 24 h,48 h and 72 h,respectively.The method was to detect the inhibitory effect of HCPT and CIK cells on liver cancer cells.The optimal concentration of HCPT and the optimal target ratio of CIK cells were screened.After 24 h,48 h and 72 h of intervention in liver cancer BEL-7402 cells,apoptosis was detected by Annexin V/PI double staining.Results HCPT(optimal concentration 0.5 mg/L)and CIK cells(optimum target ratio 20∶1)inhibited the growth of hepatocellular carcinoma cell line BEL-7402 at different times,and the combined application had synergistic effect.The anticancer effect was significantly different(P<0.05).Conclusion The treatment of HCPT and CIK cells has a significant inhibitory effect on hepatocellular carcinoma BEL-7402 cells.The combination of the two has a synergistic effect and can induce apoptosis.
