归脾汤加减方免疫调节作用研究 被引量:6

摘要 目的建立免疫抑制的小鼠模型,探讨归脾汤的组方加减对免疫抑制小鼠的免疫调节作用。方法选用昆明种小鼠,随机分成对照组、模型组和归脾汤加减方低、中、高剂量组,用环磷酰胺(CYP)诱导的方法,建立免疫抑制的小鼠模型,持续灌胃30 d,计算小鼠免疫器官(脾、胸腺)脏器指数、在显微镜下计数外周血白细胞数、用酶联免疫吸附法检测小鼠血清中肿瘤坏死因子-α(TNF-α)、免疫球蛋白G(Ig G)和干扰素-γ(IFN-γ)水平,检测脾淋巴细胞增殖能力与凋亡率和巨噬细胞吞噬功能。结果成功建立免疫抑制的小鼠模型。归脾汤组方加减提取物能够增加环磷酰胺导致的免疫抑制小鼠睥和胸腺的脏器指数,增添小鼠外周血白细胞数量,提升肿瘤坏死因子-α、免疫球蛋白G和干扰素-γ水平,增强淋巴细胞增殖的细胞活力,抑制淋巴细胞的各期凋亡率和总凋亡率并且增强小鼠巨噬细胞吞噬功能。结论环磷酰胺诱导法能够有效建立小鼠免疫抑制模型,归脾汤加减方能够较全面的从固有免疫、体液免疫等方面调节环磷酰胺导致的免疫抑制小鼠免疫功能低下作用。 Objective In order to establish a mouse immunosuppressive model and investigate the immunomodulatory effects of Guipi decoction on mice. Methods Kunming mice were used to establish a mouse immunosuppressive model by cyclophosphamide induction method, which was randomly divided into control group, model group and Guipi decoction low, medium and high dose groups, continuous intragastric administration. 30 days, then test the immune organ index, peripheral blood leukocyte count, TNF-α, IFN-γ and IgG levels, spleen lymphocyte proliferation ability and apoptotic rate and mouse carbon clearance ability and to study the related mechanism of Guipi decoction on the immunity of mice. Results The mouse immunosuppressive model was successfully established. Guipi decoction can increase the immune organ index, increase the number of peripheral blood leukocytes, increase the levels of TNF-α, IFN-γ and IgG, and enhance the phagocytosis of mouse macrophages in immunosuppressive mice induced by cyclophosphamide. Function, enhance lymphocyte proliferation, inhibit lymphocyte apoptosis rate and total apoptosis rate. Conclusion The cyclophosphamide induction method can effectively establish a mouse immunosuppressive model, and Guipi decoction can comprehensively regulate the immune function of immunosuppressed mice induced by cyclophosphamide from innate immunity and humoral immunity.
