摘要
目的研究刺蒺藜-杜仲药对对自发性高血压大鼠(SHR)左室肥厚的影响及其可能作用机制。方法将21只8周龄雄性SHR随机分为刺蒺藜-杜仲药对组(22.12 g·kg^-1)、缬沙坦组(13.35 mg·kg^-1)和SHR模型组(0.5%CMC)各7只,另将7只同周龄雄性WKY大鼠作为正常组(0.5%CMC)。各组灌胃给药,连续10周。大鼠尾动脉血压计测量大鼠收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP)。超声心动图分别测量室间隔厚度(IVST)与左室后壁厚度(LVPWT)、左室舒张末期容积(EDV)、左室收缩末期容积(ESV)、左室短轴缩短分数(FS)、射血分数(EF)、每搏输出量(SV),计算左室质量(LVM)及左室质量指数(LVMI)。取全心称重,计算心脏指数(CI)。左室心肌组织HE染色,病理图像分析系统进行心肌细胞横径(CTD)测量和形态学观察。Western Blot检测心肌组织PI3K和PKC的蛋白表达。结果与正常组比较,模型组大鼠SBP、DBP、MAP明显升高(P<0.01),IVST、LVPWT明显增厚(P<0.05),CI、LVMI、CTD显著增加(P<0.05),心肌组织形态学有明显改变,心肌组织PI3K和PKC的蛋白表达水平明显增高(P<0.05);与模型组比较,刺蒺藜-杜仲组和缬沙坦组大鼠SBP、DBP、MAP均明显下降(P<0.05),IVST、LVPWT明显变薄(P<0.05),CI、LVMI、CTD有所降低(P<0.05),心肌组织形态学有显著改善,心肌组织PI3K和PKC的蛋白表达水平明显降低(P<0.05)。结论刺蒺藜-杜仲药对具有逆转SHR左室肥厚的作用,其机制可能与降低心肌组织PI3K/PKC信号通路的活性有关。
Objective To investigate the effect of couplet medicines of Tribulus Terrestris L-Eucommia Ulmoides against left ventricular hypertrophy(LVH)in spontaneous hypertensive rats(SHR)and the possible mechanism.Method Twenty-one 8-week-old male SHRs were randomly divided to the couplet medicines of Tribulus Terrestris L-Eucommia Ulmoides group(22.12 g·kg^-1),the valsartan group(13.35 mg·kg^-1)and the SHR model group(0.5%CMC).In addition,seven 8-week-old male Wistar Kyoto(WKY)rats were included into the normal group(0.5%CMC).All of them were orally administered with drugs for 10 weeks.The rat tail sphygmomanometer was used to measure the systolic blood pressure(SBP),diastolic blood pressure(DBP)and mean arterial pressure(MAP)of rat caudal arteries.The echocardiography was used to measure the interventricular septal thickness(IVST),left ventricular posterior wall thickness(LVPWT),left ventricular end-diastolic volume(EDV),left ventricular end-systolic volume(ESV),left ventricular fractional shortening(FS),ejection fraction(EF),stroke volume(SV)and calculate left ventricular mass(LVM)and left ventricular mass index(LVMI).The cardiac index(CI)was calculated by weighting the whole heart.The pathological image analysis system was used to measure the cardiomyocytes transection diameter(CTD)and observe the myocardial histopathology by hematoxylin and eosin(HE)staining.The expressions of PI3 K and PKC were observed by Western blot.Result Compared with the normal group,the SHR model group showed significant increased in the SBP,DBP and MAP(P<0.01),obvious growths in IVST and LVPWT(P<0.05),remarkable increase in CI,LVMI and CTD(P<0.05),significant deterioration in the pathological changes of left ventricular myocardium,and notable increases in the levels of protein expressions of PI3 K and PKC(P<0.05).Compared with the SHR model group,the couplet medicines of Tribulus Terrestris L-Eucommia Ulmoides group and the valsartan group showed significant decrease in SBP,DBP and MAP(P<0.05),remarkable reduction in IVST and LVPWT(P<0.05),significant fall in CI,LVMI and CTD(P<0.05),obvious amelioration in the pathological changes of left ventricular myocardium,and notable decrease in the levels of protein expressions of PI3 K and PKC(P<0.05).Conclusion Couplet medicines of Tribulus Terrestris L-Eucommia Ulmoides have the effect on reversing LVH in SHR.The possible mechanism may be correlated with the down-regulation of PI3 K/PKC pathway.
