摘要
淫羊藿苷是淫羊藿的主要活性成分之一,具有免疫调节、抗炎、抗衰老、抗肿瘤等活性,可改善骨质疏松、阿尔兹海默病、帕金森、脑缺血、动脉粥样硬化、风湿病等多种疾病的症状。其药理活性包括:促进成骨细胞的生成和活化,抑制破骨细胞的形成和功能缓解骨质疏松;抑制β-淀粉样蛋白的产生、tau蛋白的过度磷酸化及多巴胺含量的降低以保护神经系统;通过保护心肌细胞、增加心肌细胞数量、改善内皮功能障碍保护心血管系统;促进生殖器官的功能;通过调节T细胞平衡调节免疫系统。目前,对于淫羊藿苷的研究仍有以下不足:其药理活性与多种信号通路有关,但如何调控这些信号分子、直接效应分子尚不明确;对其药理活性的研究多为单一靶点、单一途径,说服力不强;与他药配伍增效减毒的作用有待挖掘。今后的研究首先应借助激动剂、拮抗剂和基因编辑,明确淫羊藿苷调控信号通路与相应药理作用的关系,揭示通路变化的上游及下游效应分子;其次,联合采用表面等离子体共振、配体-受体结合实验、靶分子共定位、质谱等方法论证其多途径、多靶点作用,增加说服力;再次,结合中医药理论与现代分子生物学技术,对淫羊藿苷与他药配伍的药理作用进行深入探讨。
Icariin is one of the main active ingredients of Yinyanghuo( epimedium). It has immunomodulatory,anti-inflammatory,anti-aging,anti-tumor and other activities. It can improve osteoporosis,Alzheimer’s disease,Parkinson’s,and cerebral ischemia.,Atherosclerosis,rheumatism and other diseases. Its pharmacological activities include: promoting the formation and activation of osteoblasts,inhibiting the formation and function of osteoclasts to alleviate osteoporosis;inhibiting the production of β-amyloid protein,the excessive phosphorylation of tau protein and the reduction of dopamine content to protect Nervous system;protect the cardiovascular system by protecting myocardial cells,increasing the number of myocardial cells,improving endothelial dysfunction;promoting the function of reproductive organs;regulating the immune system by regulating T cell balance. At present,the research on icariin still has the following deficiencies: its pharmacological activity is related to multiple signaling pathways,but how to regulate these signal molecules and direct effector molecules is not clear;researches on its pharmacological activity are mostly single targets,The single approach is not convincing;the effect of compatibility with other medicines to increase effectiveness and reduce toxicity needs to be explored. Future research should first use agonists,antagonists,and gene editing to clarify the relationship between icariin-regulated signaling pathways and corresponding pharmacological effects,and reveal upstream and downstream effector molecules of pathway changes;second,the combined use of surface plasmon resonance and ligands-Receptor binding experiments,target molecule co-localization,mass spectrometry and other methods to demonstrate its multi-path,multi-target effects,increase persuasiveness;again,combining traditional Chinese medicine theory and modern molecular biology technology,compatibility of icariin with other drugs The pharmacological effects of this are discussed in depth.
