摘要
目的建立沙棘配方颗粒的质量标准。方法采用TLC法对沙棘配方颗粒剂进行定性鉴别;采用醇溶热浸法测定沙棘配方颗粒的浸出物;采用紫外光谱法测定沙棘配方颗粒中总黄酮的含量;采用HPLC法测定沙棘配方颗粒中异鼠李素、山柰素、槲皮素的含量,并研究沙棘配方颗粒的指纹图谱。结果沙棘配方颗粒的TLC鉴别方法专属性强,斑点清晰。芦丁0~47.2169 mg·L-1与峰面积呈良好的线性关系(r2=0.9990),平均加样回收率为98.90%(RSD=0.76%,n=9);槲皮素0~110.650 mg·L-1与峰面积呈良好的线性关系(r2=0.9999),平均加样回收率为99.93%(RSD=1.55%,n=9);山柰素0~112.880 mg·L-1与峰面积呈良好的线性关系(r2=0.9999),平均加样回收率为100.05%(RSD=2.65%,n=9);异鼠李素0~170.000 mg·L-1与峰面积呈良好的线性关系(r2=0.9999),平均加样回收率为98.54%(RSD=2.30%,n=9)。指纹图谱研究显示:10批次沙棘配方颗粒的相似度良好,均大于99.8%。结论所建薄层色谱方法显色清晰,适用性强;各成分的含量测定方法准确可行,重复性好,浸出物和有效成分的含量限定合理,可有效控制沙棘配方颗粒的质量。
OBJECTIVE To establish the quality standard of the Sea buckthorn dispensing granules(SBDG).METHODS The SBDG were identified by TLC.The content of the extract was determined by alcohol-dissolving hot-dip method.The content of total flavonoids in SBDG was determined by UV spectrophotometry and the content of isorhamnetin,kaempferol and quercetin was determined by HPLC.Meanwhile,the fingerprints were established in which isorhamnetin,kaempferol and quercetin characteristic peaks were identified.RESULTS TLC spots of SBDG get clear focus and good separation.The rutin(0-47.2169 mg·L-1),quercetin(0-110.650 mg·L -1).Kaempferol(0-112.880 mg·L -1) and isorhamnetin(0-170.000 mg·L -1) showed good linear relationships and the average recovery rates were 98.90%(RSD=0.76%,n=9),99.93%(RSD=1.55%,n=9),100.05%(RSD=2.65%,n=9) and 98.54%(RSD=2.30%,n=9),respectively.The fingerprint results showed that the 10 batches of Sea buckthorn dispensing granules had good similarity which is all above 99.8%.CONCLUSION The established TLC method has clear color and strong applicability.The detection method of each component is accurate and feasible with good repeatability.The content limitations of extract and active ingredient are rational,which can effectively control the quality of Sea buckthorn dispensing granules.
