白藜芦醇联合5-Fu对高糖诱导的视网膜色素上皮细胞氧化损伤的保护作用 被引量:1

The Protective Effect of Resveratrol and 5-Fu on High Glucose-induced Oxidative Damage in Human Retinal Pigment Epithelial Cells

摘要 目的探讨白藜芦醇(Res)联合5-氟尿嘧啶(5-Fu)对高浓度葡萄糖(Glu)诱导的人视网膜色素上皮细胞ARPE-19氧化损伤的作用及其可能机制。方法ARPE-19细胞分为五组:正常对照组(5.5mmol/L Glu),高糖组(30mmol/L Glu),Res组(30mmol/L Glu+10μmol/L Res),Res组联合5-Fu组(30mmol/L Glu+10μmol/L Res+0.6μg/mL 5-Fu),5-Fu组(5-Fu,30mmol/L Glu+0.6μg/mL 5-Fu)。CCK-8法检测Res对ARPE-19细胞增殖的影响,确定Res对ARPE-19细胞最佳作用时间和最佳作用浓度。CCK-8法检测Res联合5-Fu对ARPE-19细胞增殖的影响。检测SOD、MDA、GSH-Px的活性。结果Res对ARPE-19细胞的最佳作用时间为48h,最佳作用浓度为10μmol/L。Res组能增强人视网膜色素上皮细胞活力(P<0.05)。Res联合5-Fu组增强人视网膜色素上皮细胞细胞活力作用较Res组更加明显(P<0.05)。Res组能显著增加ARPE-19细胞SOD、GSH-Px的活性,降低MDA活性(P<0.05)。Res联合5-Fu组使ARPE-19细胞SOD和GSH-Px升高更明显、MDA减少更显著(P<0.05)。结论Res联合5-Fu可通过调节SOD、GSH-Px、MDA等抗氧化因子的活性保护视网膜色素上皮细胞。 Objective To explore the protective effect of resveratrol and 5-Fu on high glucose-induced oxidative damage of ARPE-19 in human retinal pigment epithelial cell line ARPE-19 and its possible mechanism.Methods ARPE-19 cells were divided into five groups including the normal control group(5.5 mmol/L glucose),high glucose group(30 mmol/L glucose),resveratrol group(30 mmol/L glucose+10μmol/L Res),resveratrol group combined with 5-Fu group(30 mmol/L glucose+10μmol/L Res+0.6μg/mL 5-Fu),5-Fu group(30 mmol/L glucose+0.6μg/mL 5-Fu).The CCK-8 method is used to detect the effect of resveratrol on the proliferation of ARPE-19 cells.The optimal action time and optimal concentration of resveratrol on ARPE-19 cells were determined.The effect of resveratrol combined with 5-Fu on the proliferation of ARPE-19 cells was detected by CCK-8 method.The activity of SOD,MDA,GSH-Px was tested according to the kit instructions.Results The optimol actiom time and optimal concentration of resveratrol on ARPE-19 cells was 48 h and 10μmol/L.Resveratrol can enhance the viability of human retinal pigment epithelial cells(P<0.05).Resveratrol combined with 5-Fu enhances the viability of human retinal pigment epithelial cells more significantly than the resveratrol group(P<0.05).Resveratrol can significantly increase the activity of SOD,GSH-Px and reduce MDA activity.Resveratrol combined with 5-Fu significantly increased the activity of SOD and GSH-Px,and significantly reduced the activity of MDA(P<0.05).Conclusion Resveratrol combined with 5-Fu protects retinal pigment epithelium cells by modulating the activity of antioxidants such as SOD,GSH-Px,and MDA.
