摘要
目的测定马钱子生品、砂烫炮制品和牛奶炮制品中马钱子碱和士的宁含量,及其毒性研究。方法采用HPLC法测定马钱子生品、砂烫炮制品和牛奶炮制品中马钱子碱和士的宁含量,通过小鼠急性毒性实验测定马钱子生品、马钱子砂烫炮制品及马钱子牛奶炮制品的半数致死量(LD50)。结果马钱子生品中马钱子碱和士的宁的含量分别为(0.037±0.014)mg/g和(0.934±0.254)mg/g,LD50为1.9737 mg/kg体质量。马钱子砂烫炮制品中马钱子碱和士的宁的含量分别为(0.016±0.005)mg/g和(0.237±0.098)mg/g。马钱子牛奶炮制品中马钱子碱和士的宁的含量分别为(0.011±0.006)mg/g和(0.191±0.048)mg/g。马钱子砂烫炮制品和牛奶炮制品按最大给药剂量(2.70 mg/kg体质量)给药后,小鼠均无出现死亡。结论砂烫、牛奶炮制能够降低马钱子生品中马钱子碱和士的宁的含量,提高用药的安全性。
Objective To determine the contents of brucine and strychnine in Strychnos nux-vomica L.raw products,processed products of sand scalding and processed products of milk,and to study their toxicity.MethodsThe contents of brucine and strychnine in the nux vomica,nux vomica processed products and milk processed products were determined by HPLC,and the LD50 of the nux vomica,nux vomica processed products and nux vomica processed products were determined by acute toxicity test in mice.Res-ultsThe contents of brucine and strychnine in raw nux vomica were(0.037±0.014)mg/g and(0.934±0.254)mg/g respectively,and LD50 was 1.9737 mg/kg.The contents of brucine and strychnine in the processed products were(0.016±0.005)mg/g and(0.237±0.098)mg/g respectively.The contents of brucine and strychnine in processed milk were(0.011±0.006)mg/g and(0.191±0.048)mg/g respectively.There was no death in mice after the maximum dosage(2.70 mg/kg body weight)of nux vomica and milk processed products.ConclusionSand scalding and milk processing can reduce the contents of brucine and strychnine in raw nux vomica and improve the safety of drug use.
